B lymphocyte is the primary immunocompetent cell. After stimulated with antigen and other stimuli, B cell undergo activation, proliferation and differentiation process and then play multi-function in immune response. So, the B-cell activation is a very important premise for exertion its role. Studies showed that resting B cell enter activated state accompanied with some expression variation of many genes, in which some are up-regulated while others are down-regulated. It is the interaction of these genes's product that lead to the B cell activation. Unfortunately, the gene regulation in such a process still blur for us till now. Cloning and functional analysis of the B cell activation-related gene will not only help to understand the molecular basis of normal B cell growth and development, but also to elucidate the pathogenesis of B cell-derived immune disorders such as leukemia, autoimmune diseases and lymphoma. Therefore, to find novel B cell activation-associated gene is one of hot study fields in immunology.In the pre-phase of our work, 62 different expressed cDNA fragments (ESTs) have been isolated after using differential display reversal PCR (DDRT-PCR) to compare the gene expression difference between resting and activated B cells. In order to clone the full-length of EST's corresponding gene, we adopt the strategy as bellows: firstly, we use northern-blotting to eliminate the false-positive ESTs.
