With the development of science and the intersections of varies subjects' theories and methods, the development and maturity of molecular biology is fastened. Molecular biology theories are enriched not only by recent years' new technologies, new methods, new theories and new ideas in this field but also by the fulfillment of Human Genomic Project and the achievements of the following Human Postgenomic Project research. At the same time, molecular biology is also facing the challenge of decisive problems of the new century. That is, future molecular biology will probe into the basic problems of inheritance, development, evolution and function adjustment not from the separate but from the genomic structure and functions viewpoint. Therefore, based on the achievements of Human Genomic Project, researching extensively and thoroughly on genome and post-genome of agricultural economic animals has great immediate and historical significance on probing into the inheritance, development, evolution, the expression of genes, adjustment mechanism and their genetic breeding of fine livestock animal and poultry from molecular level.Researching on the tissues of rat, mouse, human-being, dog, carp and bullfrog, the cloning and sequencing of 26 hormone categories and their relative protein gene segments has been carried on. At the same time, probed with some of their gene segments, using the methods of hybridization, immunohistochemistry, in situ hybridization and Northern Blot, basic research was done on the location, determination of the nature, fixed quantity, special expression, time and space expression of mRNA in different tissues. The consistence of cloned 26 genes' agarose gel electrophoresis result and the sequencing result showed that the cloned gene is the target gene. The Northern Blot hybridization and the numerical value of hybridization zone's densitometric scan absorbance radiated from development was measured. The result shows that mRNA of experiment group and comparison group in different tissues and same tissue have expressive differences. The result of in situ hybridization shows that the mRNA expressive extent of rats' Cerebral artery membrane Bkca passageway under slowly enhanced alcoholic effect related with controlled conditions. Bkca passageway effected critically in the product of cerebrospinal fluid when alcoholic exists. PACAP not only take part in the production of steroid in granular cell but also promote ovulation in interstitial cell. In order to ovulate, PACAP adjust gonad stimulating hormone by combining with its receptor at least in egg follicle's granular cell before ovulation. The object of this experiment is to lay a foundation of experimental material, methods, technology and theory for the future research on structural genome, functional genome and genetic breeding of fine livestock animal and poultry.
