Studies On Function Factor For Lycium Barbarum Polysaccharide (LBP): Characteristics And Biological Activity Of LBP-X

It is widely recognized that Lycium barbarum L. (LBL. ) play multiple roles in pharmacological and biological functions as a tra-ditional Chinese herbal medicine and also a kind of food. The studies were performed on characteristics and biological activity in purified fraction of Lycium barbarum polysaccharide (LBP - X) in order to ensure its believable and scientific degree as a functional factor of LBL. in the third generation functional food on molecule level. The main results are as follows:1. Characteristics of functional factor LBP-X Isolation and purity Extracting LBP from LBL. and separated LBP -X by ion-exchange cellulose and gel chromatography which is a complexe of heteropolysaccharide-protein. Effects on food propeties The results showed that LBP -X is not effect propeties of food in different pH values and concentration while it was added into food. The condition was given that further study on steady-state of food functional factor LBP - X.2. Biological activity of functional factor LBP-X : Action of decreasing blood glucose The results of medical sequential trial indicated that LBP - X 10mg/(kg d) had obvous actions in reducing blood glucose (decreasing values:8. 12 mmol/L in empty stomach, 14. 13 mmol/L after a meal 2hr)and the curve of glucose tolerance of alloxan-induced diabetic rabbits which did not affect the blood glucose of normal mice. Using the matched pair sequential trials compare the effect of LBP-X on reducing blood glu-cose with LBP and LBL. that the effect of LBP -X is the best a-mong three (decreasing blood glucose values, LBP: 4. 09 and 6. 83 mmol/L, LBL. :3. 98 and 5. 55 mmol/L). The effective rate of reducing blood glucose is 100% and the rate of false positive 1%. Function of decreasing blood lipids The results of medical sequential trial also showed that the effect of LBP -X 10mg/(kg d) is remarkable on decreasing the content of tolal cholesterol and triglceride(TC:4. 27 mmol/L and TC:3. 50mmol/L) and increasing high-density lipoprotein in serum of alloxam-induced hyperiipi-demia rabbits (decreasing serum lipids, LBP: TC 3. 82 and TG 1. 69 mmol/L, LBL. :TC 3. 39 and TG 2. 77 mmol/L). LBP-X is not effect the serum lipide of normal rabbits which the effective rate of decreasing serum lipids is 100% and the rate of false positive 5%. Action of Antifatigue Five different does of LBP - X (5mg/(kg d), 10mg/(kg d), 20mg/(kg d), 50mg/(kg d), 100mg/(kg . d) and LBP 10mg/(kg . d) were applied to mice to study their effects on antifatigue and the relatibinship between dose and respose, and compare the effect of LBP-X on antifatigue with LBP. The results showed a remarkable effect on enhancing the reserves of muscle and liver glycogen, increasing latic dehydrogenase (LDH) activity before exercise and after swimming 90 min and 150min, decreasing the content of raise of blood urea nitrogen (BUN) and accelerating the clear rate of BUN after strenuous exercise. The reserves increasing rate of muscle and liver glecogen (MG &. LG) are test group from 14. 24 to 64. 93%(MG) and 40. 00 ~83. 33%(LG), control group 5. 80 (MG) and 8. 86% (LG) a-mong them LBP -X 10mg/(kg . d) equal 64. 93%(MG) and 83. 33%(LG), LBP 10mg/(kg . d) are 28. 82%(MG) and 65. 71% (LG) separately. Befor exercise increasing rate of LDH are from 9. 52% to 15. 25% and after swimming 90 min 10. 00 to 19. 31% in which LBP-X 10mg/(kg . d) are 15. 26% and 19/31%, LBP10mg/(kg . d) equal 10. 96% and 12. 86% separately. Raising content of BUN from 8. 91 to 23. 78% among which LBP -X 10mg/ (kg . d) make 8. 91% and LBP. 10mg/(kg . d) 11. 08%. Effects of LBP-X 10mg/(kg . d) was the best among the five doses of LBP - X. Comparing affects of LBP - X on antifatigue with control group (normal saline) were significance difference (P<0. 01) and comparing with positive control group LBP 10mg/(kg . d) which effects of low doses (5mg/(kg . d), 10mg/(kg . d) and 20mg/(kg . d)) on antifatigue were much better than LBP(P0. 05)...
