Monolithic silica column, a novel type chromatographic column totally different from traditional column in concept, has been got much attention in recent years. But the crack and deformation of column bed during the preparation of monolithic silica column were the serious problem. In order to avoid the problems, N,N-dimethylformamide was used as a drying control agent to control drying rate. With this approach, the drying time was shortened greatly and the deformation was restrained when the wet column bed was pre-heated before drying in order to strengthen the strength of silica skeleton.Different concentration ammonium was used porogen solvent to fabricating the different size mesopore and the fabricating mesopore's methods at high and low temperature were investigated for the different monolithic column beds in diameter.
