Purpose: with the rapid development of modern agriculture and industry, the highly materialcivilization is brought to human society, and meantime the survival circumstance of mankind isfacing increasing challenges. The Over release of chemical substance has polluted theenvironment severely, more and more evidence shows that many synthetic material whichrelease to the environment has the function of interfering the endocrine system of human andwildlife, and influencing the growth, reproduction and the behavior of organism themselves andtheir descendants. Recently some endocrine-active compounds in the environment were havingsignificant adverse effects on public and environmental health, which disturbs the homeostasisof endocrine system, so called endocrine-disrupting chemicals (EDCs). Since the publicattention has been focus on the threats causing by the environmental pollution, it is urgently tofind out an effective assessments of toxity of environmental endocrine disruptors.Method: the serum vitellogenin (VTG) of bullfrog (Rana catesbeiana) and goldfish(Carassius auratus) shows a well dependability with the calcium concentration, so calciumconcentration was used to evaluate estrogenic activity. after exposure to different dosage of E214d, determined the serum VTG concentration of male and female goldfish by ELISA method.and determined the serum calcium concentration by atomic absorption spectroscopy. Determinethe VTG level of same month male and female bullfrog which come from water sourcedownstream of luo-tong River in Korea and agriculture land, Bussan, during May to August.Study the relevance of induced VTG and genital glands development: Gonadosomatic index(GSI) (weight of genital gland/weight of body×100%). Primary culture the liver cells of youngfish of goldfish. 17β-dihydrofolliculin and chromium were used to induce VTG and HSP-70experiment, the concentration of VTG was determined by ELISA. HSP-70 was determined bywestern blot, the results was analyzed with grey scale analysis by digital gel analysis system.Result: after exposed to different level of E2 14d. the ELISA determined concentration ofVTG was significantly increased(P<0.01).comparing with control group, and the serum calciumconcentration shows and dosage dependent manner(R2=0.768): the VTG inside agriculture landbullfrog are much higher (p<0.05)than that from water source downstream, while GSI are lower(p<0.05) than their counterparts For the frog from the water source downstream ofluo-tong River in Korea. GSI of July are higher than that of May and June. And VTG of Juneis higher than May and July; the goldfish in the downstream show a low GSI and high VTG,and bisexual reproduction was found, alter exposed to different level of 17β-dihydrofolliculinand chromium, the VTG and HSP-70 could be found in the liver cells of young fish. The 48hinduced level is greater than that of 24h.it expressed most under E2500ng/L+cdS0ug/L andcd100ug/L(p<0.01).Conclusion: the concentration of calcium in goldfish serum reflects the concentration ofVTG indirectly. So it could use to determine the interfering degree of endocrine-disruptingchemicals on fish. The downstream of luootong River in Korea and agriculture land werepolluted in some extent; endocrine-disrupting chemicals such as 17β-dihydrofolliculin andchromium play a role in inducing VTG and HSP-70 in the liver cells of young fish of goldfish:HSP-70 is an effective, specific and sensitive biological warning marker for early stageenvironment pollution.
