Blue honeysuckle(Lom'cera Caerulea L.) is a kind of wild berry plant, mainly distributed in Russian, China, Japan and North America. As having quite high nutrients value in its fruits, it obtained high recognition by some countries. Some excellent strains have been cultivated in Russian and Japan as well as been planted artificially.Though we have an abundant wild resource in China, there were few studies on its breeding and resource. The study on germplasm resource of Lonicera Caerulea L. was not much made abroard and many problems should be resolved. Upon these plants, some germplasm resources were collected at home and abroard, and their morphology, palynology, cytology, growth and development habit, fruit economy characteristics were studied. The extraction methods for DNA of Lonicera Caerulea L. and reaction system of RAPD were also discussed in the project. And it was the first time to study the genetic diversity, relationship and classification of Lonicera Caerulea L on wild resource in Northeast of China and subsection using RAPD. So the germplasm resource of Lonicera Caerulea L can be evaluated and will provide the basic study for breeding. The main conclusion were as follows: 1 The biological characteristic of wild blue honeysuckle in northeast region1.1 Morphological studiesThe exterior morphological characteristic of wild Lonicera Caerulea L in northeast region varied much among inter-populations and intra-populations. There is only a remarkable difference between Mohe population and Wangqing, Yabuli, Yuchi, Boli, Wuying population. The general morphological variation is not remarkable among other populations.1.2 Palynological studiesThe pollen shape of six populations were all spheroid. (28.51-40.93) 34.16x28.84 (23.78-32.52) urn. Equator axis was 1.16-1.20 and with spine enation on ektexine. The difference of pollen shape between populations was not significant; decorative pattern were the same. This showed that the relationship among the populations in northeast region was much close.1.3 Cytological studiesThe base numbers of chromosome were X=9, and diploid, the whole number were 18. All of them were made up of middle centromere. The karyotype formula in Mohe population was2n=2x=l8=l6M+2SM and that in other five populations was 2n=2x=l8=l2M+6SM. The karyotype type of all populations were 2C, belonging to evolution type. 1.4 Growth and developing characteristicThe seeds of Lonicera Caerulea L. had an obvious light accelerated germination characteristic. The optimal germination temperature was 20 C~25 C, which could make seeds germinate without sand-stored treatment.The rapid growing stage of young shoots in Harbin was for the first twenty days of May. The growing curve was in"S" shape. The whole growing stage was 156-163 days.2 The extraction of DNA and RAPD conditionsUsing modified SDS methods high quality DNA was extracted. The optimal reaction condition was as follows: pre-denaturation at 94 C for 4mins; denaturation at 94"C for Imin; annealing at 36 C for 40s, extending at 72 C for 1 min, the whole reaction repeats 40 cycles with a last extension at 72 C forSmin.3 The genetic diversity and realtionship between blue honeysuckle populations in Northeast regionPerformed RAPD using 27 random primers, the total of 186 loci were detected and among them 121 loci were polymorphoric, accounted for 65.05% of total loci. The mean polymorphoric percentage of each population was 38.89%. Most difference was among wild populations in the total genetic difference.The relationship between populaions was much close. According to genetic similarity, the 6 populaitons could be clustered into 3 groups. The Boli, Wangqing, Yabuli and Yuchi populations belonged to the first group, Wuying populaiton to the second group and Mohe population to the third group.
