Recently DNA vaccines have been widely used in HIV vaccines. The poor immunogenicity is an innative deficiency for DNA vaccine. One approach to boost the immunogenicity of DNA vaccine consists of co-delivery of plasimids encoding some natural immune stimulators. Cholera toxin is enterotoxin made by Vibrio Cholerae. Its holotoxin, A subunit and B subunit can all be used as mucosal adjuvants.In this proposition, we tried construct some recombinant plasmids (along with some recombinant TTV) that co-express CTB and some special antigen,then use this as DNA vaccines with adjuvants. Moreover, we will observe the parental and mucosal adjuvanticity of these plasmids.1. CTB and antigens co-expressing DNA vaccines’immunogenicity via intramuscular inoculationThe TRIVN-CTB DNA vaccine raised stronger cellular and humoral immune response than the TRTVN DNA vaccine. DNA vaccine co-expressing CTB and TRIVN as adjuvant woked well.2. CTB and antigen co-express DNA vaccines’immunogenicity via mucosal administration After confirming the adjuvanticity of the DNA vaccine via intramuscular inoculation, we decided to study the mucosal adjuvanticity of the DNA vaccine.The results showed that OVA-CTB rTTV boosting raised more rigorous antigen-specific T cell immune response in both system and mucosal lymphoid tissues (proximal, distal and mesenteric lymph nodes) than OVA rTTV boosting. But there was no significant difference about the antigen-specific antibody titer in serum, bronchical lavage fluid and vaginal lavage fluid.In brief, this thesis tried to construct CTB and special antigen co-expressing DNA vaccines, then used them as vaccines with adjuvants in both systemic and mucosal inoculation routes. It is a new idea of adjuvants development or DNA vaccine modification.
