The rats were randomly divided into normal control group,chronic pelvic inflammatory disease model group,Fuyanning soup low,medium and high dose group and Kangfuyan capsule group,enzyme-linked immunosorbent assay(ELISA) to detect the rats factor serum levels of inflammatory cells, including interleukin-8(IL-8), interleukin-4(IL-4). After the rat uterus morphology between ELISA method to detect the rat uterine tissue adhesions related cytokines in cell adhesion molecule-1(ICAM-1),transforming growth factor-β(TGF-β)expression,fluorescence in real time quantitative PCR(Real Time-PCR) detection of epidermal growth factor(EGFm RNA) expression; application blot(Western Blot) to detect the expression of uterine tissue of rats in each group of NF-KB.Results:1. The naked eye Normal control group:normal uterine size and shape,and the surrounding tissue adhesion does not occur,the endometrium and no congestion Edema. Pelvic inflammatory disease model group:pelvic inflammatory exudate visible,uterus distal fallopian tubes,ovaries wrapped into a package block,and fiber-like adhesions surrounding tissue,uterine swelling, tortuous,water-like changes were seen uterine fluid uterus less flexible,tough texture. After the Chinese intervention,uterus, fallopian tubes and ovarian mass was reduced to varying degrees,uterine shape gradually returned to normal,with the high-dose group Fuyanning most significant changes in the uterus and the distal tubal adhesions ovarian mass is not obvious,only a small number of scattered fibronectin,like changes in the water disappeared uterus,uterine tissue elasticity close to normal. 2. Pathological morphology by light microscopy Normal control group: uterine mucosa, muscular and serosa stratification clearly structured endometrial glands,no edema,no inflammatory exudate; muscle fiber texture clear; serous smooth,no congestion,edema and inflammatory infiltration out. Pelvic inflammatory disease model group:uterine tissue structural disorder,unclear boundaries,tissue congestion,edema,lesions involving the endometrium, myometrium,serosa,lymphocytes and plasma cells mainly diffuse chronic inflammatory cell infiltration,some glandular sac hyperplasia and fibrosis seen. The lesion is particularly evident within the film,part of the model also shows granulation tissue-like changes. After a traditional Chinese medicine,the treatment groups compared with chronic inflammatory changes in model group,particularly in the high-dose group Fuyanning significantly reduce the uterus hierarchical organization structure more clearly,only a very small amount of scattered chronic inflammatory cells present in the intimal layer,endometrial glands structured,no change in fibrosis. 3. ELISA detection of rat serum IL-8 and IL-4 results Serum content model group and IL-8 were significantly higher than the normal group,the difference was significant(P <0.01); after the Chinese intervention showed different degrees of decline,each Chinese intervention group compared with model group had highly significant difference(P <0.01),in which the high-dose group close to normal levels, no significant difference between two groups(P> 0.05); Fuyanning, the high dose group was significantly lower(P representing Kangfuyan group <0.01),Fuyanning low-dose group than in the low Kangfuyan group,but there was no significant difference(P> 0.05). Significantly lower serum content of IL-4 in the model group than the control group(P <0.01), each Chinese intervention group had different degrees, compared with model group with a significant difference(P <0.05 or P <0.01),but there is still a significant difference compared with the normal group(P <0.01); Fuyanning low dose,Kangfuyan group was significantly lower than Fuyanning high-dose group(P <0.01); Fuyanning dose group than in the significantly increased Kangfuyan group(P <0.01),Fuyanning low-dose group and Kangfuyan group,no significant difference(P> 0.05). 4. ELISA detection of uterine tissue in ICAM-1,TGF-β results Uterine tissue model group ICAM-1 content than normal group significant difference was highly significant(P <0.01); each medicine intervention group compared with model group were decreased to varying degrees,there was a significant difference(P <0.01); Fuyanning high dose group close to normal levels,with no significant difference(P> 0.05); no significant difference Kangfuyan capsule group and Fuyanning low-dose group,and in the high-dose group there was a significant difference(P <0.01). RT-PCR detection of uterine tissue in ICAM-1m RNA, TGF-βm RNA, EGFm RNA results Model of uterine tissue more significant increase in ICAM-1m RNA with the normal group, the difference was significant(P <0.01), Fuyanning, the high-dose group compared with model group, the difference was significant(P <0.01);no high-dose group and the normal group significant difference(P> 0.05); there was no significant difference(P> 0.05)levofloxacin group and model group. Model of uterine tissue more significant increase in TGF-βm RNA expression of the normal group,the difference was significant(P <0.01); Fuyanning,a more significant decline in the high-dose group and model group,the difference was significant(P <0.05 or P <0.01);there was no significant difference(P> 0.05)levofloxacin group and model group. Uterine tissue expression of EGFm RNA,the normal group was significantly higher in the model group, the difference was significant(P <0.01); Fuyanning soup,high-dose group and model group there was a significant difference(P <0.01); no significant difference in the high-dose group and the normal group(P> 0.05); there was no significant difference(P> 0.05)levofloxacin group and the untreated group. 6. Western Blot detection of uterine tissue results in NF-KB Uterine tissue expression of NF-KB,the model group and the normal group,were significantly increased,the difference was significant(P <0.01); Fuyanning high dose compared with model group,significantly decreased,the difference highly significant(P <0.01); no significant difference high-dose group and the normal group(P> 0.05); there was no significant difference between levofloxacin group and model group(P> 0.05).Conclusion:1. Apply the mixed bacteria and methods intramuscular injection of dexamethasone intrauterine successfully copied chronic pelvic inflammatory disease(deficiency syndrome)in animal models of disease and syndrome. 2. Chronic pelvic inflammatory disease(deficiency syndrome)in rats there are also related to abnormal expression of cytokines. 3. Fuyanning soup can improve the dexamethasone-induced deficiency syndrome abnormal behavior of rats to learn the model. 4. Fuyanning soup can regulate the expression of cytokines and nuclear factor NF-KB to regulate chronic pelvic inflammatory disease(deficiency syndrome)immune function in rats,and thus significantly improve the chronic pelvic inflammatory tissue injury and fibrosis. 5. Fuyanning soup is an effective prescription treatment of chronic pelvic inflammatory disease(deficiency syndrome),and has broad application prospects. |
