Text one:Changes and Significance of Blood Stasis Type of Arteriosclerosis Obliteransassociated Factors in Serum Metabolites and Inflammatory ReactionPurpose: Study of blood stasis type of arteriosclerosis obliterans(ASO) changes and significance of related factors in serum metabolites and inflammatory reaction.Material and method:15 cases of patients with blood stasis type ASO, 10 healthy control subjects enzyme linked immunosorbent assay(ELISA) double antibody sandwich methodin detection of serum VEGF, TGF-β1 and IL-1β level, gas chromatography-mass spectrometry(GC/MS) technique based on metabonomic analysis of endogenous metabolites in serum in the methodological change.Results: Compared with the control group, the content of serum IL-1 increased significantly in the serum in ASO group, there were very significant differences(P < 0.01); VEGF,TGF-β1 levels increased slightly, and there were no significant differences in the metabolism of carbohydrates, amino acids; water, fatty acids in serum in ASO group of average happened obvious change.Conclusion: Closely related to changes in the incidence of type ASO blood stasis andinflammation and serum endogenous metabolites..Text two: Experimental study on the relationship between the vascular endothelial cell injury, the change of the serum endogenous metabolites and pathogenesis of arteriosclerosis obliteransPurpose: To discuss the relationship between the vascular endothelial cell injury, the change of the serum endogenous metabolites and pathogenesis of arteriosclerosis obliterans(ASO).Material and method:ASO models were constructed with the method of high-fat diet plus intimal injury, at 8 weeks after operation, the arterial morphology was observed using light microscopy, using percoll density gradient centrifugation method to determe the number of circulating endothelial cells(CEC), serum endothelin-1(ET-1) and Nitric oxide(NO) levels were detected using enzyme linked immunosorbent assay(ELISA) double antibody sandwich assay, the change of the endogenous metabolites in serum were analyzed with the metabonomics method-based gas chromatography-mass spectrometry(GC/MS) technology.Results: Rat models of ASO were established successfully, the number of CEC in ASO rat group peripheral blood increased significantly, the serum ET-1 level were significantly higher, and the level of NO was significantly less than normal control after operation, and there was significant difference(P<0.01). Compared to normal control group, metabolism of the sugars, amino acids, fatty acids in serum of ASO rats were significantly changed.Conclusion: Vascular endothelial cell injury and the change of serum endogenous metabolites mediated ASO, protect the vascular endothelial function, adjust the balance of the endogenous metabolites should be a important therapeutic targets of ASO.Text three:Blood stasis syndrome serum of arteriosclerosis obliterans on vascular endothelial cell injury and regulation of Taohong Siwu DecoctionPurpose: To discuss the blood stasis syndrome serum of arteriosclerosis obliterans on vascular endothelial cell injury and regulation of Taohong Siwu Decoction.Material and method:To establish umbilical vein endothelial cell culture system, endothelial cell injury model was made by ASO blood stasis syndrome serum, the drug serum of Taohong Siwu Decoction low, medium, and high concentration were added respectively, endothelial cell proliferation activity was observed by MTT method, ultrastructure of endothelial cells were observed with transmission electron microscope, the change of intracellular calcium ion concentration and the cytoskeleton were observed with laser confocal microscope, and Enzyme-linked immunosorbent(ELISA) to observe ET, NO and TGF-beta 1 content in each group endothelial cell culture supernatant.Results:In ASO blood stasis syndrome serum group, endothelial cell proliferation activity decreased, the cell structure destroyed, Calcium ion concentration increased, ET and TGF-beta 1 content increased significantly, ET/NO ratio imbalance. After incubating With Taohong Siwu Decoction drug serum, endothelial cell proliferation activity and damage of cell structure obviously improved. TGF-beta 1 levels drop, ET/NO ratio returns to normal.Conclusion: These findings indicated that the main mechanism of Taohong Siwu Decoction in the treatment of blood stasis syndrome ASO for improving the injury of vascular endothelial cells and secretion dysfunction.
