Objective To study the different expression of micro RNA between the plasma of asymptomatic hyperuricemia and the healthy control using gene chip technology. Make the target gene predication to the differently expressed micro RNA and explore the action mechanism of micro RNA in patients with hyperuricemia.Methods 1.Four asymptomatic hyperuricemia and four healthy control were recruited,whose RNA in peripheral plasma were extracted by method of Trizol. The micro RNA expression profile was tested and the expressed micro RNA was screened out by micro RNA miroarray(Exiqon TM company,Danmark). 2.Real time RT-PCR was used to verify the results between 50 asymptomatic hyperuricemia and 50 healthy control. And the clinical data of 50 cases of hyperuricemia group and 50 cases of control group were analyzed. 3.The target genes of these micro RNAs were predicted through searching and summarizing the miranda database, mi RBase database and targetcsan database, and the target genes were analyzed by GO and Pathway.Results 1. MicroRNA microarray expression showed that there were 263 microRNA in the hyperuricemia group and control group. Five obviously up-regulated and six obviously down-regulated micro RNAs in the hyperuricemia group were identified. 2.Using RT-PCR technology, the expression of mi R-519d-3p was up-regulated in the hyperuricemia group, the difference was statistically significant(P<0.01); the expression of mi R-30c-5p and mi R-374a-5p were down regulated, and the difference was statistically significant(P<0.01). The results were consistent with micro RNA microarray. 3.Through the prediction and functional analysis of the micro RNA gene, the expression of micro RNA was found to play its role in the TGF- pathway, PI3K-Akt signaling pathway, endocytosis and Notch signaling pathway.Conclusion Micro RNA microarray technology is an effective method for high throughput analysis of micro RNA expression profile. Compared with the normal control group, the expression of differently micro RNA in the hyperuricemia group may become a new molecular biology marker and may be involved in the pathogenesis of hyperuricemia.
