On the basis of the theories of TCM about emotion caused diseases and modem medicine and physiological-psychological medicine, using physiological and psychological experiment methods, to research influences of emotion causes on type 2 diabetes and the mechanisms; and to observe influences of emotion causes on the rats behavior and the macroscopic and microscopic regulatory action to rats of decoction Xinshen No.5. The research results add to discover the mechanism of emotion caused diseases and its biological basis.The animal models of emotion caused type 2 diabetes were made by abdominal injecting little dosage of STZ and imposing experiment emotional stimulation (rotation, restraint and crowding) to rats simultaneously. The experiment animals were divided into five groups. Each group was checked of weight and blood sugar level every two weeks and killed after 6 weeks. Then chemical changes in rats' hypothalamus-pituitary-adrenal axis under emotion stimulation were examined by methods of radioimmunology, immunohisto-chemisry and in situ hybrid; the transcription level of insulin receptor mRNA was examined by RT-PCR; the adrenaline and noradrenaline level in blood were examined by HPLC. And the influence of emotional stimulation to rats' action was evaluated by the open-field testo The multi-path and multi-link regulatory functions of decoction were observed in synchronism. Result1.Apparent and persistent blood sugar level increasing and blood insulin level decreasing were observed in the rats injected with STZ under emotional stimulation. The differece was observed between the groups. Compared with the other groups, the decogroups1 blood sugar level is considerably lower and blood insulin level is higher.2.Emotional stimulation can increase the transcription level of c-fosmRNA and CRFmRNA and the translation level of FOS protein and the blood ACTH and cortisol level. Each level of rats of STZ plus stimulation group were considerably higher than other groups (p<0.01 or 0.05). And the decogroups' were the lowest.3.Emotional stimulation can increase rat' blood adrenaline level. STZ plus stimulation group have the highest level and were different to stimulation orSTZ groups. The adrenaline level of decogroup is the lowest among allgroups.There were no apparent difference of noradrenaline level between eachgroup.But there was increasing tendency of noradrenaline level in STZ plusstimulation group. 4.Emotional stimulation can increase the transcriptional level of mRNA of liverinsulin receptor. The STZ plus stimulation group have the highest level(p<0.01or 0.05) and the decogroup have the lowest level. 5.Emotional stimulation can depress the rats' action especially the STZ plusstimulation group.Compared with the STZ plus stimulation group, the changesof decogroup is considerably light(p<0.01 or 0.05). Conclusion 1 .The animal model was made by emotional stimulation and STZ methods.Theexperiment results prove that the emotional stimulations can aggravate thepersistent carbohydrate metabolism disorder in type 2 diabetes model rats theninduce or aggravate type 2 diabetes. 2.Emotional stimulation can induce type 2 diabetes through multi-path and onmulti-target. The most coherent paths are hypothalamus-pituitary-adrenal glandaxis and vegetative nerve-adrenal gland medulla.The increasing of insulinreceptor mRNA level is the periphery moleculor pathology mechanism. 3.Emotional stimulation can induce rats' abnormal action which express chieflyas depressed action. That reflects that Qi disorder maybe the results of lost ofcoordination between mentality and physiology. 4.Shenxin No. 5 may interfere the activation of hypothalamus-pituitary-adrenalaxis and vegetative nerve-adrenal gland medulla then to lighten bad negativeinfluence of emotional stimulation on type 2 diabetes.
