Hepatic chirrosis is a kind of common disease which is severely impairing the health of Chinese people. In the part of the disease itself, liver chirrosis is a chronic, proceeding and distributed lesion in liver. Fibrosis is the inevitable mediate stage of varied chronic liver disease to chirrosis. So far, great developments have been made in the etiology, pathogenesis and diagnosis hepatic chirrosis, and especially in the treatment of the disease. In order to further propagate the superiority of Chinese herbs in the treatment of hepatic fibrosis, and deeply recognize the practical efficacy and their mechanisms, we designed and executed the following studys on the basis of two kinds of fibrotic model of rats.1 The time-effect study of two animal models of rats in hepatic fibrosis1.1 Objectives To analyze the differences between the two kinds of hepatic fibrotic animal models, which are CC14 and pig serum methods, in time-effect and pathologic feature in the course of development of liver fibrosis.1.2 Methods Using CC14 and PS ten-week methods to make rat models of liver fibrosis. Forty male SD rats, weight 180-220g, were randomly divided into two groups. Rats in group one were given 40%CC14 oil solution at the dose of 2ml/kg, subcutaneous injection twice weekly. Rats in group two were given pig serum which was obtained from original mix blood of adult pigs also at dose of 2ml/kg, intraperitoneal injection twiceweekly. The features of pathologic development of liver fibrosis at 6, 10, 14 and 20 weeks of model making were observed respectively by Masson stain method, and the presentations were analyzed by computerized image analysis system.1.3 Results In CC14-method group, fibrotic pseudolobulars developed at the 6th week of animal model making, became typical at the 10th week, and got tranquilization thereafter, with ballooning degeneration of liver cells outstanding. On the contrary in PS-method group, fibrosis occurred late in 10 weeks during model making, and developed from 10 weeks through 14 weeks though the model making had stopped; meanwhile ballooning degeneration was not presented.1.4 Conclusions There was indeed something different in the development of liver fibrosis time-effect relation and pathologic feature between the two groups of model making. This indicated different mechanisms were referred in the pathogenesis of liver fibrosis. The two methods of model making should be combined in the coming animal study of liver fibrosis; the prophylactic and treatment studies by anti-fibrotic drugs should last no less than 14 weeks, and should begin 4 weeks ahead of the beginning of and the end of model making.2 Heteroserum-induced liver fibrosis of rats is modulated by a -SMA and TIMP-1 positive cells2.1 Objectives The activated HSC, which is featured by the expression of a -SMA, plays a key role in the pathogenesis of liver chirrosis. TIMP-1, another very important factor which is also expressed by HSC, down-modulate the activity of fibrolysis. In this study, we aimed to look at the modulating effect of a -SMA positive cells in the development of liver fibrosis.2.2 Methods2.2.1 Animal model Twenty eight SD rats were randomly divided into two groups, with 20 rats in model group and 8 in control. Rats in modelgroup were given PS intraperitoneally, 2ml each, twice weekly for 1 Oweeks. Rats in control were given saline at the same method as model group. At the 6th, 10th, 14th and 20th week after the beginning of model making, 5 rats were sacrificed respectively in model group, and 2 rats in control. The liver was removed and disposed as following managements. 2.2.2 Pathology observation The liver tissue was dehydrated, enbeded in paraffine wax for pathological section.
