Part IThe Experimental Study of the FFXHHG in Rehabilitation of the Wounded Soft TissueObjective To investigate the mechanism of FFXHHG which could promote the rehabilitation of the wounded tissue by regulating the fibroblast apoptosis and the genetic expression of the basic fibroblast growth factor(bFGF) in wounded tissue.Method According to the method of building models by beating, sixty wistar mouse got acute soft tissue's lesion. Then they were divided randomly into three groups: group A, used nothing; group B, healed with positive pharmaceutical; group C, coped with FFXHHG. Both group A and group B were control groups, while group C was experimental group. The rehabilitation of the wounded tissue was observed respectively at 2, 4, 7 and 10 days since the mouse were dealt with pharmaceutical, in accordance with the histomorphology, the immunohistology and the RT-PCR analysis of genetic expression of bFGF. Results Observed in histomorphology: myofibers were all turgid, broken and solvable and there were inflammatory cellular infiltration in three groups, butgroup A was prior to the other two. After being cured, the capillary hyperplasia in group B and group C was more evident than that in group A, more granulation tissue grew and local tissue was repaired well. TUNEL detective result displayed that the fibroblast in wounded tissue was expressed positively in group B and C, especially in group C, while in group A, the fibroblast apoptosis became weakly positive. S-P detective method demonstrated that: In wounded tissue, the positive signals of alpha-smooth muscle actin(a-SM actin) and bFGF lied mostly in cellular membrane were manifested violently in group B and C, while that in group A was normal. With the healing going on, the positive signals in group B and C abated step by step, on the contrary the positive signals in group A became stronger and stronger at 10 days. RT-PCR analysis of the genetic expression of bFGF showed: At 2 days the product rate of PCR in group C was almost the same as which in B (P>0.05), but it was obviously different from the rate in group A (PO.01). At 4 days, the difference of the PCR produce rate among three groups had remarkable significance, but the difference between group B and A got little important (p<0.05). With the time didn't affect the statistical results (p>0.05). At 7 and 10 days, the electrophoretogram displayed: the PCR product decreased greatly and there were almost nothing in three groups. All of the date demonstrated that: FFXHHG and Wan Hua You(WHY) could enhance the levels of the genetic expression of bFGF in wounded tissue, while the level in FFXHHG decreased more slowly than the other two. Conclusion FFXHHG could promote the fibroblast apoptosis and regulate the bFGF genetic expression in wounded tissue during the rehabilitation. The protein synthesis in fibroblast and extracellular matrix was stimulated directly by FFXHHG because of bFGF, and formed collagen fibril later. FFXHHG could be combined with the relevant receptors, induce endotheliocyte migration, improve the capillary hyperplasia and formed fibrocyte. All these might be the1999primary mechanism that the Chinese Herbal Medicine, which could activate blood circulation to dissipate blood stasis, could accelerate the rehabilitation of the wounded soft tissue.Part IIThe Clinical Study of FFXHHG for the Rehabilitation of Wounded Soft TissueObjective To evaluate the clinical effect, the coefficient of resistance and the adverse reaction of FFXHHG used to cope with the wounded soft tissue. Method According to the method of being compared with each other in random, eighty patients with acute soft tissue lesion were researched in clinic. Results In FFXHHG group, the total effective rate was 87,27%, the pain, swelling, ecchymosis and function disorder were improved. But in the pain and function disorder had more significant efficacy than that of controls (P<0.05).
