1.PurposeIdiopathic thrombocytopenic purpura (ITP) is a common hemorrhagic disease in clinic. Its morbidit reason is not clear until today. This experiment investigated the function of immunological organs in the ITP from the aspect of the immunological organs' structure (such as thymus,spleen).We detected the change of the immunocytes and cytokines and the internal relationship between them and the expression of the apoptosis and apoptosis protein of the spleen and thymus lymphocytes in order to explain the function of them in the episode of the ITP. We also examined the count and function of the blood platelet, observed the character of the change of the megakaryocyte in bone marrow to understand the relationship between them and the episode of the ITP, then to know the effective mechanism of the ZIDIANKELI.2.Method 2.1Experimental ResearchWe established the BALB/c mouse model of chronic ITP by using the method of immunization( abdominal cavity injection APS on the BALB/c mouse ). Div-iding the mouse into different groups randomly according to the count of the blood platelet, we explored the effective mechanism of the ZIDIANKELI to the ITP model mice from four different levels: wholism,organ,cell and molecule.We researched the thymus and the spleen's structure changes by the method of HE staining to explain the relationship between them and the ITP. We detected the changes of the T-cell subgroup by the method of flow cytometry, examined the change of the IL-2, IL-6, IL-8 by the method of radio-immunity to definite the function of them in the episode of the ITP. We also detected the apoptosis rate of the thymus and spleen lymphocytes and the expression of the Fas and Bcl-2 protein with the TUNEL method and the immunohistochem istry method to understand the relationship between them and the ITP.2.2Theory ResearchWe expounded the immune episode mechanism and the treatment of the clinical drug by reviewing all the Traditional Chinese Medicine records and Modern Medicine records. In the research of the general view of Traditional Chinese Medicine, we summarized the idea of the etiological factors and pathology of ITP in Traditional Chinese Medicine systematically and explained the present state of the ITP Traditional Chinese Medicine treatment and the progress of the Traditional Chinese Medicine experimental research. Meanwhile, we discussed the founded background of ZIDIANKELI, and we treated the ITP according to the idea "the origin of the disease is the Qi-Deficiency, Blood Stasis run through the course of the disease" and make the principle of the treatment "strengthening Qi and tonifying the deficiency, removing the blood stasis and producing the fresh blood.3.Result 3.1Experimental Research3.1.1The effects of ITP model mice's general state and swimming time by the treatment of ZIDIANKELIAfter the fifth day of establishing the model, the model group appeared following symptoms: activity reduced, liking to gather together, piloerection, no sheen of the hair, swimming time reduction. To compare with the normal group, it is has statistical significance.P<0.05.After 48h APS rejection to the mouse, some ecchymosis and petechia appeared on the abdominal and tail parts. All the experimental groups can improve the symptoms in different degree. In general the effects of the big dosage of ZIDIANKELI group is the best.3.1.2The effects of the ZIDIANKELI to the immunological organ of the ITP model miceThe mice of the model group have following changes: the enlargement of the spleen, thymus atrophy. To compare with the normal group, it is has statistical significance.P<0.05, The result showed: The effects of the big dosage of ZIDIANKELI group is the best in general, it can improve the immunological organ obviously.3.1.3The effects of ZIDIANKELI to the peripheral blood platelet count of the ITP model miceThe blood platelet count of the model group decreased obviously, to compare with the normal group, it is has marked statistical significance.
