The study of correlation of syndrome with treatment is a weak point of syndrome study which has been becoming a hot point in diagnostics of Chinese medicine in recent years. Stress is nonspecific and protective adaptation mechanism of human body. It also results in the change of homeostasis even diseases. Stress reaction of human body can originate from many factors which are likely to be somatic, psychological, social and cultural. Many systems are involved in stress which causes the changes of histology, physiology and biochemistry with which neuroendocrine and immunity system is closely related. Some relative research indicates that 75-90 percent epidemic diseases have something to do with stress which its action is gradually paid attention. Especially over last decades, research about stress predominates over the total health of human body.Objective of this study is as follows: ①Make the rats model of chronic immobilization stress and evaluate it; ②Systematically explore the pathological and physiological bases from the theoretical and experimental study; ③Comparably study the action mechanism of anti-stress of compositus of Chinese herbs Xiaoyaosan, Sijunzitang and Jinkuishenqiwan; ④Explore and discuss the correlation of syndrome with treatment of rats model of chronic immobilization stress.This thesis is one main part of national natural science fund assistance item(№:30000216)and Huo Yingdong enducational fund assistance item (№:81037).It can be divided into two part:theoretical research and experimental research.Theoretical researchThe following contents were mainly explored in theoretical research. ①concept and stage of stress; ② knowledge about stressors and action of Liver, Spleen and Kidney on stress in Chinese medicine and main pathogenesis of chronic immobilization stress; ③method of making animal model of stress; ④relation of stress with neuro-endocrine-immunity network; ⑤relation of stress with neuropeptide; ⑥relation of stress with trace element Zinc and Selenium. Experimental researchThe first experiment: make and evaluate the rats model of chronic immobilization stress and effect of Compositus of Chinese herbs on model rats.Objectives of this experiment: ①Explore behavioral change of rats under the condition of chronic immobilization stress; ②Explore the effect of three recipes of Chinese herbs on behavioral change of rats of chronic immobilization stress; ③Survey changes of fodder intake and bodyweight of rats under chronic immobilization condition; ④Compare the effect of different treatment on fodder intake and body weight of chronic immobilization stress rats;⑤ Evaluate the rats model of chronic immobilization stress. Sixty male rats were randomly divided into six groups and 10 rats in each group: the normal control group, model group of 7 days, model group of 21 days, the group treating with Sijunzitang or Xiaoyaosan or Jinkuishenqiwan( respectively simplified as the Sijunzitang group, the Xiaoyaosan group, the Jinkuishenqiwan group). Recipes used in the experiment were respectively from Formularies of the Bureau of People's Welfare Pharmacies (Xiaoyaosan: Beichaihu 30g, Danggui 30g, Baishao 30g, Baizhu 30g, Fuling 30g, Gancao fried with honey 15g, Shengjiang 10g, Bohe 10g; Sijunzitang: Dangshen 30g, Baizhu 15g, Fuling 15g, Gancao fried with honey 10g) and Synopsis of Prescriptions of the Golden Chamber (Jinkuishenqiwan: processed Dihuang 60g, Shanyao 30g, Shanhzuyi 10g, Zexie 15g, Fuling 15g, Mudanpi 15g, Guizhi 10g, processed Fuzi 10g). Rats model of chronic immobilization stress was made by binding rats on the specific bound frame three hours a day for 21 days (7 days in group B). Chinese herbs were made into powder by certain procedures for usages. Xiaoyaosan, Sijunzitang and Jinkuishenqiwan were respectively injected into the stomach of rats in the treatment groups and same volume of distilled water was used for rats in the other group one hour before rats were bound.
