Part 1 Experimental Research on Taohongsiwu Decoction Intervening Blood Vessel Regeneration of CallusObjective: To explore the mechanism that Taohongsiwu Decoction (THSWD) intervenes blood vessel regeneration and reconstruction of bone stump and consequently promotes the healing of fracture.Methods: In the complete random projects, after the operative fracture experiment was executed on the rabbits, the rabbits were given orally with THSWD (observation group), Shangkediedapian (SKDDP) (Positive control group) and distilled water (model control group) respectively. Moreover, the other group of rabbits (normal group) reared at the same time was chosen as anormal contrast. At the 7th, 15th, 21st and 35th day after treatment, we took the X-ray films on the extremities of fracture and tissue of bone stump (callus) in every group above. Then we analyzed blood vessel regeneration and observed the dynamic change of VEGF protein and mRNA expression.Results: Based on the X-ray films, we found that blood swelling in bone stump in THSWD group and SKDDP group was absorbed quickly in early phase, and furthermore, the growth callus was apparently better than in the model group. Moreover, we also found that the observation group was the best among three groups. This indicated that THSWD and SKDDP significantly improved the growth of callus and promoted the healing of fracture. THSWD made the healing process of fracture into the stage of new bone formation in advance. In the micrangium morphosis of callus, at the 7th, 15th day after treatment, micrangium morphosis of callus multiplied extensively, and the quantity ofmicrangium in the observation group and control group was significantly more than that in the model group (P<0.01). However, from the 21st day to the 35th day after treatment, the quantity of blood vessel of callus decreased gradually and the morphosis trended to normal bone tissue, and regenerating blood vessel reached the reforming process, and the phase of reconstruction and remodeling of the vessels of callus tissue advanced more in the observation group and the control group than in the model group. Nevertheless the observation group was the best. In VEGF protein and mRNA of callus, after treatment, the quantity of VEGF expression increased at the 7th and 15th day and reached the peak value. In the two periods the expression quantity was more in the observation group and the control group than in the model group (P<0.01), and the quantity of VEGF expression decreased more slowly in the observation group and the control group than in the model group at the 21st, 35th day, and the trend was the most obvious in the observation group. However, it was not more significant than the former two periods (p>0.05). EVGF expression quantity was more in the observation group than in the control group and the model group at the 35th day (p<0.01), and the expression level of VEGF protein was identical with that of mRNA. It indicated that THSWD and SKDDP could improve the expression level of VEGF protein and mRNA of bone stump and retard the decreasing trend of expression after the peak value. The effect of THSWD was better. The higher concentrations of expression were maintained in the region of bone stump.Conclusion: THSWD improved the transcription levels of VEGF, mRNA of callus tissue in bone stump and promoted VEGF protein expression. It could maintain higher expression environment in the region of bone stump where it stimulates proliferation of regenerated blood vessel of callus in early phase and accelerates the reconstruction of micrangium in callus in the later phase. It could accelerate blood swelling absorption and promote the growth and remodeling of the callus, then promo the healing of fracture.
