Cornus officinalis Sieb., listed in 2000 Chinese Pharmacopoeia, is firstly recorded in "Shennong's Herbal". The dried fruit of this plant has been claimed to replenish liver and kidney arrest seminal enission in Chinese medicine system. This plant is widely used in Chinese folk medcine with various pharmacological properties. Recent studies demonstrated that this medicine had an unique therapeutic effectiveness in treating diabetic angiopathy. Therefore, the purpose of this study was to develop new drugs of Cornus officinalis Sieb which is more effectively against diabetic angiopathy.1. To ensure the quality of new drug, the origin and use of this plant was investigated and confirmed by making a comprehensive survey of the production place - xixia county of Henan province. The results showed that this place has the advantage of sufficient supply of Cornus officinalis Sieb with stable quality, which could meet current requirement for new drug development.2. In view of the nationwide complicated classifications for Cornus officinalis Sieb. a more detailed classification study was performed. The results showed that the existing general concept for the classification of this plant was reasonable through field survey and literature analysis3. By orthogonal experimental design, the method for the extraction and purification of iridoid and in the active parts of this medicine was optimized. A series of technical parameters was set up accordingly. The key procedure in the preparation of this new drug was also quantitatively monitored so as to assure the detectable content in iridoid and .... was above 45% with HPLC, 70% with UV4. To control the quality of this new agent more effectively, the fingerprint of Cornus officinalis Sieb and its semi-product was firstly established on the basis of fixed species, production place, pickup season and process method. The precision, reproducibility and stability of the method for fingerprint was also evaluated. The common and typical chromatography pattern produced with HPLC were established through the determination for up to 10 batches of Cornus officinalis Sieb and its semi-product. Meanwhile, 4 indicative components, morrom'side, loganin, sweroside and ursolic acid were identified in the chromatography.5. Several new procedures which are different from that of Chinese Pharmacopoeia for quality control was established in this study. For example, the simultaneous determination of two more active components in iridoid with HPLC, the separation and determination of two structurally similar chemicals, ursolic acid and oleanolic acid with HPLC, thus provide a new methodfor the quality control of this medicine.6. 10 chemicals were separated from the active parts of this medicine through systematical separation procedure. 6 out of them were identified, In addition, 3 chemicals-morroniside, loganin and sweroside were prepared and purified by prepared HPLC as standards.7. The pharmacological results revealed that both the active parts of iridoids and triterpenes acid had effects on decreasing blood glucose, though the joint use showed the better results.In sum, systematical studies for Cornus officinalis Sieb and its agent were performed in this paper from the aspects of basic research to practical application, the results of which would provide an more fundamental basis for the development of new drugs in treating diabetic angiopathy, also produce a new insight into the ongoing study for Cornus officinalis Sieb.
