Glioma is a kind of malign tumors which were created in encephalic epithelium . Its incidence accounts for 44% au brain tumors. Glioma has many characters including quick growing up, bigger sockage and higher recommitting rate than that of other tumors, which severly affect human health. Curing glioma has many traditional integrated measures which includes sperotion, radiotherapy and chemotherapy, etc.Recently, some new methods are coming, such as immunetherapy and genetherapy. Traditional therapies have many serious side effects and new therapy methods have not maturely formed. So, Lest side effects and best curing effects is important measure for curing glioma.Recently, men has knew tumor apoptosis which is a complex course affected by many factors. Research of tumor apoptosis and its control mechanisms have become focus in tumor treatment fields. Many research confirmed that gliomas have apoptosis phenomena. The aim of this research is to find a kind of drug which can promott glioma apoptosis and not affect normal cells. Thulium is a kind of minim elements in human body. Lacl3 is a lighter compound in thulium. Toxicologically research showed that Lacl3 haven't posion side effects on normal human body. But thulium compound has many biological activities, Which can change membrane fluidity and ATP enzyme activation and ion transition and mitosis, DNA synthesis, thinks thulium used as drugs is a new research field in 1987. Now, thulium compound have bellowed effect as clinical drugs. First, resistiny flammatron and sterilize effects. Second, decreasing blood sugar effects. Third, promoting tissue grouing. Fouth, resisting tumor and promoting tumor apoptosis effects. Research has been confirmed that it can promote stomach cancer and lung cancer apoptosis. This research obserre that resist proliferation and promote apoptosis on brain glioma SHG-44. With Lacl3 and explore its molecular mechanisms. Otherwise, Our conntry have much thulium. Its reserves account for 80% all world reserves. So it has good applying prospective.This research including, first, culturing human brain glioma SHG-44, and forming good cell vines. Second, different concentration (0.01,0.1 and 100 mmol/L) thulium drug administed SHG-44, and we observe apoptosis changes with morphology methods. MTT method, electron microscope, flow cytometry method and TUNEL method, ets. Third, we observed PTEN,Bcl-2 and Bax protein change in SHG-44 ceus with immunhistochemistry methods. Fourth, we also observed its related gene mRNA changes with in situ hybridization.Our results showed, First, compared with control group, every treatment groups SHG-44 have some sow proliferation and apoptosis change . Its cell arrange sparse, most cells showed cell volume shrinage, chromotin aggregation observed microscope. Second, with concentration of Lacl3 increasing, OD volue decreast and inhibition rate increase. Third, ultrastructure change of SHG-44 showed than cell volume reducing, nuclear chromation sparre and forming nuclear frogmentation and apoptotic bodys with electron microscope methods. Fourth, SHG-44 apoptotic percentage increasing with flow cytometry methods. Fifth, More cells have PTE,Bcl-2 and Bax gene ecpressions in SHG-44 with in situ hybridzation .So, we think thulium compound Lacl3 have signification proaportotic effects on human brain glioma. Increasing PTEN and Bax gene expression and decling Bcl-2 gene expression probably have important effect on its proapoptotic effect.
