The essential hypertension is a kind of popular disease throught the world. there are over 10% people who suffer hypertension in our Country. But there are fewer people know they suffer the disease and Even less people who take the medicine and control the hypertension. Why? To answer the question, one of the most important reason of problem is the details and the mechanisms are not completely understood until now. The most advanced development about hypertension is insulin resistance theory. Some clinical investigations of the hypertension indicate the insulin sensitivity is about 40% less amount hypertensive subjects and the insulin level is 25~40% higher than normal subjects. Others, including outside and domestic researchers, have been trying to use insulin sensilizers to administer on hypertensive patients and their relative. Those researchers got some very surprising results: insulin sensilizers could reduce the high blood pressure by attenuate the insulin resistance and hyperinsulinemia. There are more researchers use insulin sensilizers, such as Rosiglitazone, on the animal models of hypertension to explore the effect and the mechanisms. And they are even want to evaluate the protective effects on the cardiovascular system. Nowadays, some problems are much concerned in the field of research. 1,Rosiglitazone could reduce the blood pressure of spontenously hypertensive rats (SHR) or not? Until now there are only a couple of experiment was performed on spontenously hypertensive rats. Because most of the experiment were performed on the vary kinds of secondary animal models. SHR rats is a kind of model whose genetic backgrounds and the course of high blood pressure is more assemble of the hypertension of human beings. It means the results from SHR experiment may be more repeatable on patients. Rosiglitazone is a kind of thiazilididione compounds which are more effective and less side effects .We are going to use rosiglitazone on SHR rats to evaluate its effect on the high blood pressure. And nifedipine was used as the positive control medicine. 2,The protective effects of Rosiglitazone on cardiovascular system: The cardiovascular complications of the hypertension are very common and serious. They are the high reason of morbidity and mortality of hypertensive patients. Any anti-hypertensive agents without cardiovascular protective effect could not be used on patients. So to estimate the cardiovascular protective effects is very important. There are some clinical investigates show Rosiglitazone has some effects of cardiovascular protection such as improve heart function, reduce the infracted size and prevent restenosis after coronary stent. Unfortunately, there is lack of the experiment of morphologic research on SHR. We are going to use light and electronic microscope to examine the hearts and aortas of SHR to identify the cardiovascular protection of Rosiglitazone. 3,The mechanisms of Rosiglitazone lowers the blood pressure and prevents the cardiovascular complications: If Rosiglitazone could lower the blood pressure and prevent the cardiovascular complication, how does it do so? There are some reports concerning about Rosiglitazone on kidney reabsorbing Na + and water, sympathetic nervous system activity, renin-angiotensin-aldosteron system, endothelial function, leptin, and some other factors. This field of research is one of the most advanced part of the life science all over the world. We are going to evaluate the angiotensin II receptor type 1 (AT1) and mRNA by means of the situ hybridization and the immunohistochemical method. Materials and Method: 1,Animals and how to feed the animals: 36 SHR rats were all male, birthage 7~8 wks body weight 130~160g/each. All rats were randomly divided into three groups, each group has 12 rats equally. Every 6 rats were feed in a cage and all rats were offered the some chew food and water.
