The dried roots of Polygonum cuspidatum Sieb. et Zucc is a well-known traditional drugin China. It has been widely used for the treatment of suppurative dermatitis, gonorrhea, favusathlete's and ambustion in Chinese folk medicine. Stilbene and anthraquinone is the dominantchemical composition of this plant. In recent years, stilbene-resveratrol is attentioned widely.Modern research showed that resveratrol has multi-functions including the antioxidant activity,antibacterial, preventing cance and inhibiting platelet aggregation et al. Anthraquinone alsohas much biological activity. For better use of Polygonum cuspidatum, it is importance tostudy its biological activities, extraction and purification.Coloumn chromatography was used for further separation of stilbene and anthraquinonein Polygonum cuspidatum, and NMR was applied to identification of the compound obtained.They were piceid, resveratrol, emodin-8-O-β-D-glucoside, physcion, emodin and rhein.Analysis of stilbene and anthraquinone in Polygonum cuspidatum extracts can be performedby reversed-phase high-performance liquid chromatography couped with electrosprayionization mass spectrometry (RP-HPLC/ESI-MS). By RP-HPLC/ESI-MS, three stilbene andseven anthraquinone identified in crude Polygonum cuspidatum extraction. There were onestilben and two anthraquinone have not been reported in Polygonum cuspidatum.According spectral characteristics of resveratrol (glycoside), the first derivative spectrumwas established to determine the content of stilbene in Polygonum cuspidatum. This methodcould eliminate the influences of other compositions. Evaluation of the method proved that ithad good repeatability and high recovery. HPLC method was set up by optimizing thechromatograph conditions to determine the contents of resveratrol and piceid of Polygonumcuspidatum. Anthraquinone content of Polygonum cuspidatum was measured directly bydifferential chromatograph. As compared to conventional method, this method was simple andquick to operate with discarding the steps of hydrolization and extraction.The extraction and isolation of resveratrol (glycoside) and anthraquinone in Polygonumcuspidatum with reflux method were studied. The optimal condition was 80% ethanol,extraction 4 hours, 60℃ and the ratio of material to liquor (1:20) by single factor andorthogonal experiments. The optimal conditions with microwave assisted extraction (MAE)was power 240W, 70% ethanol, the ratio of material to liquor (1:8), microwave radiation 150sby means of orthogonal experiment. Comparing reflux extraction, MAE was high efficiency,energy and time saving. For isolation of resveratrol (glycoside), the adsorption and desorptionperformance of 23 kinds of macroporous adsorbent resins were studied and compared, andH103 resin was selected for its excellent adsorption and desorption properties. The effect ofconcentration, pH and work speed of the infusion on the adsorption of the H103 resin werestudied with dynamic adsorption experiment. The 0.7-1.0mg/mLL extraction solutionconcentration, PH of 4.10, work speed of 2BV/h were the optimal condition. Resveratrol(glycoside) absorbed on H103 resin was eluted with 4BV 80% ethanol in 1BV/h speed. Themethod of macroporous resin adsorption separate resveratrol(glycoside) was better thanorganic solvent extraction method. High purity resveratrol and piceid were obtained withpolymide coloumn chromatography.Linoleic acid system, red blood cell hemolysis induced by H2O2 and chemical irritantlight reaction experiments were used for comparing the antioxidant activity of resveratrol andpiceid. Resveratrol and piceid exhibited higher antioxidant activity than VC, VE, BHT andcrud extraction of Polygonum cuspidatum in the three tests. They could inhibit linoleic acidperoxidation. And they reduced red blood cell hemolysis induced by H2O2. Resveratrol andpiceid could scavenge O2-·, ·OH, H2O2 free radical.
