| Liver transplantation is an effective therapy to terminal or deteriorative liver disease. Living related liver transplantation has developed speedily with liver graft of high quality and good curative effect. The liver graft of living related liver transplantation in clinic is come from health adult. The first important thing is to ensure for the life safety of donor, at the same time to ensure for the need of liver volume of recipient so as to carry out the basic liver function. Animal with serve trauma or large operation are companied with intestinal endotoxemia (IETM). PH is a large operation and could also company with IETM. However, if IETM is actually occur after PH and what about its effects on liver damage, particularly on liver regeneration, are still not clear. To make these questions clear will provide with further experimental data for living related liver transplantation. This experimental study is composed of four parts. ①The formation and mechanism of intestinal endotoxemia by partial hepatectomy in rats. ②The effects of intestinal endotoxemia by partial hepatectomy on the shape and function and regeneration state of remnant liver in rats. ③The effects of intestinal endotoxemia by partial hepatectomy on the CD14 gene and cellular signal transduction system of remnant liver in rats. ④The effects of intestinal endotoxemia by partial hepatectomy on cytokines related liver regeneration in rats. Materials and methods One-handred and twenty Male Wister rats weighing 180~220g were randomly divided into three groups. Normal contrast(NC) group, sham operation(SO) group and PH group. PH was performed as described by Higgins and Anderson .SO was performed exactly as indicated for PH, except that the liver was only manipulated and not resected. Take NC rats as 0h time point, and take SO as well as PH rats at 6h, 12h, 24h, 36h, 48h, 72h, 120h, 168h respectively after PH as dynamic time point. For each time point indicated, six rats were used in parallel. The samples of plasma, remaining livers and small intestinal were stored for use. Related indexes were measured by biochemistry, radioimmunoassay, immunohistochemistry, histology, ELSIA, RT-PCR and FMC etc. Ruselts Part 1 The formation and mechanism of intestinal endotoxemia by partial hepatectomy in rats.Experiment 1 The dynamic changes of plasma endotoxin levels in rats after partial hepatectomy. The results showed endotoxin increased within 72h after PH, and there were two times peak value at 12h,48h.Experiment 2 The dynamic changes of Intestinal mucosa barrier observed by optics and electron microscope after partial hepatectomy in rats. The optics examination showed Intestinal mucosa appeared neutrophil margination and superficial necrosis of the villi within 24h after PH. Penetrated electron microscope examination showed the gap between Intestinal mucosa cells was increased and the Lanthanum(La) particles were observed among the Intestinal mucosa cells within 48h after PH. The histological appearance and permeability of Intestinal mucosa were normal in NC group and injured lightly at 6h in SO group. Experiment 3 The dynamic changes of plasma DAO, GLN and GS after partial hepatectomy in rats. The results showed DAO increased at 12h, 24h time point in PH rats, there were positive correlation between the levels of DAO dynamic changes and the levels of ET dynamic changes in plasma. GLN decreased at 12h after PH , GS increased during 6h~168h after PH. There were no correlation between the levels of GLN, GS dynamic changes and the levels of ET dynamic changes in plasma. Experiment 4 The dynamic changes of number and functional state of Kupffer cells in remnant liver of rats after partial hepatectomy. The results showed the number of Kupffer cells were increased and KC activation were not significant change at 24h after PH, but these were significant increased and activated largely at 48h. There were no correlation between the dynamic changes of the number of Kupffer cells in remnant liver and the dynamic changes of ET levels in plasma. The results of... |
