Heptic fibrosis(HF) is a pathologic process that every kind of disease cause liver injure and inflammation, in repair process ECM increases unusually and accumulate excessively. Light called HF, severity make hepatic lobule construction changed, formed false hepatic lobule and hepatic tuber, calling the cirrhosis. The essence is, the heptical stellate cells(HSC) are activated through regulation to cytokine and its network, hence to increase the synthesis of agent in extracellular matrix(ECM)while the degradation is relatively not enough that leads to the loss of dynamic equilibrium, and HF is therefore developed due to over-sedimentation of ECM. It's popularly believed at present that HF is a reversible while cirrhosis is irreversible.HSC is the most major cellular source arising ECM in HF. HSC activation is the central event in HF. While HSC activation, its biology characters has great change, that is proliferative, fibrogenic, and contractile with reduced vitamin A content. Therefore, HSC has been focus in the study about HF and become an important target for treatment of HF.Kan's KanXian Granules was formed by one of the five hundred famous Chinese medicines doctors Kanliangshi professor with his theory of YiYu. It has good anti-hepatic fibrosis effect. It has been used in clinic to therapy several hundred examples for several decades. It's important to study the effect and mechanism of Kan's KanXian Granules treatment hepatic fibrosis by animal and cell molecule level.This research composes of two sections as animal experience and cell culture experience.1 Animal experience1.1 Objective: To observe the effect and mechanism of Kan's KanXian Granules treatment hepatic fibrosis.1.2Methods: The hepatic fibrosis's rat was induced by carbon tetrachloride. The experimental objects were divided into five groups: normal control group, model control group, Kan's KanXian Granules vast dose group, Kan's KanXian Granules equal dose group, interferon group and Biejiaruanganpian group. The animal's estate, avoirdupois, the viscera ratio of avoirdupois, liver function, hepatic fibrosis guides, hepatic hydroxyproline(Hyp) in liver, histological changes and expression of α -smooth muscle actin( α -SMA) and transforming growth factor β 1(TGF- β l)of liver were observed.1.3Result: Kan's KanXian Granules can remarked improve the liver function, The animal's estate and avoirdupois, drop the level of hepatic fibrosis guides and Hyp, decrease the area of hepatic fibrosis and the expression of α -SMA and TGF- β 1.1.4Conclusions: Kan's KanXian Granules has reliable action in treatment hepaticfibrosis, which might be due to its protect hepatic cells, regulate the collagen , decrease the active hepatic stellate cells.2 Cell culture experience2.1 Objective: To observe the effect of the Kan's KanXian Granules serum to the multiplication and apoptosis and cell cycle of the activated HSC, and discuss the mechanism.2.2Methods: The cultural HSC T6 were added normal rat serum, Kan's KanXian Granules vast dose, Kan's KanXian Granules equal dose, Kan's KanXian Granules low dose, interferon group and Biejiaruanganpian serum to culture together. Observe the effect of the activated HSC multiplication by the method of MTT. The effect of the activated HSC apoptosis and cell cycle were analyzed by flow cytometry. Observe the effect of the activated HSC express TGF- β 1, PDGF-b, ICAM-1, Bax, P27, CyclinE by immunohistochemical methods.2.3Result: Kan's KanXian Granules serum can remarked restrain the activated HSC multiplication and promote the activated HSC apoptosis and restrain the cell cycle of HSC. Kan's KanXian Granules serum can decrease the expression of TGF- β 1 , ICAM-1 , CyclinE and increase the expression of Bax , P27.2.4Conclusions: Kan's KanXian Granules serum can remarked restrain the activated HSC multiplication may due to its decrease the expression of TGF-β1, ICAM-1.
