TGF-β signal transduction pathway has the ability to adjust cell cycle, it can inhibit growing and amplification of many cells. Studying with keratinocytes, we use Smad4, one of the TGF-β transductor, gene knock-out mice as animal model, and apply SB-431542, the inhibitor of TGF-P receptor, both in vivo and in tissue culture in vitro. With immunohistochemistry, we find that the wound healing of burn and sharp instrument injury in mice will be accelerated by inhibiting TGF-p signal transduction pathway; with XTT and flow cytometry, the function of TGF-β1 inhibiting mice kerotinocytes growing and the opposite function that SB-431542 does have been confirmed.
