| Objective: To study the effect and mechanism of gross saponins from Tribulus Terrestris L(GSTT)on myocardium ischemia/reinfusion in mice. Methods: 1. To set mice model of MI/R, and observe pathologic changes of myocardium with HE stainninng, myocardial infarction area with triphenyl tetrazolium chloride(TTC) staining, infiltration of polymorphonuclear neutrophils(PMN) in myocardium with Myeloperoxidase(MPO), and detect lactic dehydrogenase lactic dehydrogenase(LDH), creatine phosphokinase (CPK), malondialdehyde (MDA), tumor necrosis factor-a(TNF-α), interleukin-1β(IL-1β) with RIA, apoptosis rate of cardiac muscle cell with FCM, protein expression of Bax, Bcl-2, ICAM-1, NF-κB p65 with immunohistochemical method. 2. To set MI/R model of culture cardiac muscle cell, give a morphological and surviving rate observation with TEM, detect LDH, CPK, MDA, TNF-α, IL-1β with RIA in culture fluid, ICAM-1 expression in cardiac muscle cell with ELISA, and detect apoptosis rate of cardiac muscle cell with FCM, protein expression of Bax, Bcl-2, ICAM-1, NF-κB p65 with immunohistochemical method. Results: GSTT could improve myocardial pathologic and ultrastructure changes in MI/R, reduce myocardial infarction area, inhibit infiltration of PMN, reduce LDH, CPK,MDA, TNF-α, IL-1β release, inhibit apoptosis of cardiac muscle cell, down regulate Bax gene and up regulate Bcl-2 gene, and inhibit protein expression of ICAM-1, NF-κB p65. Conclusion: GSTT has an effect of protecting MI/R in mice, its mechanism might relate with improving myocardial pathologic and ultrastructure in MI/R, reducing myocardial infarction area, inhibiting infiltration of PMN, alleviate injury of free radical,... |
