Dendritic cell (DC) is the most powerful antigen-presenting cell compared with otherantigen-presenting cells. The main characteristic of the cell is that it can stimulate theoriginal type of T cell to proliferate , promote the born of CTL and Th., start theimmunity response induced by the T cell. DC is very rare in one's body which confine thestudy of it's biological character. Cord blood is resourceful , easy to collect and has weakimmune reaction. Cord blood CD34+ is more pure than marrow or peripheral blood celland has stronger proliferation which makes it the ideal source of DC.Osteosacoma is the common malign tumor. Because of the limit of chemotherapy orradiotherapy, we focus more on the immunotherapy.To investigate the feasibility of reducing the tumor antigen peptide dose and inducingthe anti-osteosarcoma immunity by dentritic cell-presenting to elucidate the characterisiticsof modifying DC by heat shock protein (Hsp70) and antigen peptide .Osteosarcoma antigenpeptide bound to Hsp70 was used to modify DC in vitro . The metabolism of the modifiedDC and the cytokine secreted thereby was tested. Then the activation of lymphocytes bythe modified DC and Hsp70 peptide was tested.. The inhibition of osteosarcoma in mice byDC injection and Hsp70-osteosarcoma peptide was tested. osteosarcoma peptide bound toHsp 70 could mature DC . Obviously anti-osteosarcoma effect could be seen when 8×104matured DC was injected in mice . we make a conclusion that DC-presenting pathway toactivate lymphocytes could kill the tumor effectively.
