Inhibition of the Recurrence of Hepatitis B Virus after IL iver Transplantation by Recombinant Adenoviral Vector Encoding Full Length Gene of HumanizedAntibody against Pres2 Antige n..Liver transplantation is now the most effective method for end-stage liver disease management. The recurrence of hepatitis B is now the major problem in liver transplantation. In this study, the recombinant adenoviral wector encoding full length gene of humanized antibody against pres2 protein of hepatitus B virus was generated. The recombinant virus was transfected into CHO cells, WRL-68 cells , L-02 cells, primary human hepatocytes,rat liver graft during cold pre servation and its efficacy in treating posttransplantation HBV recurrence was evaluat ed.. The pres2 antibody was expressed by the recombinant virus containing the whole hummanized antibody gene of pres2 antigen both in vivo and in vitro. The affinity of the amtibody for pres2 antigen was 2.16 ± 0.20 X 108L/mol.The pres2 antibody can protect: human hepatocytes from hepatitis B virus infection. This might be a new methcd- to prevent hepatitis Breinfection after liver transplantation.
