| Objective During clinical practice, it is important to explore the internal infection rule of patients' skin so as to find out how to set up an efficient bacterial barrier which prevents incision and inner incision contamination. This study is also trying to seek a more efficient bacterial test for clinical practice and laboratory tests. Method After the investigation of common bacteria on patients' skin and post-operative incision, the PCR test of 7 kinds of bacteria had been set up. At the beginning, end and every 30 minutes during the operation, samples were collected from skin, incision edge pericardium and compartment with sterilized cotton stick. Conventional bacterial culture and PCR test were performed in both animal test and clinical patients in order to evaluate the difference between traditional surgical barrier and new plastic incise drape. Results PCR test could give the result within 5 hours. The sensitivity of PCR could be up to single clone, which has a significant difference compared to conventional bacterial culture. The microorganism from patients' skin started growing one hour after the beginning and migrating deeply into the pericardium and compatment tissue of incision. The new plastic incise drape performed a significant prevention of the bacteria from growth and migration. Conclusion The growth and migration of microorganism from patients' skin are the main resource of surgical contamination. The new plastic incise drape could reduce incision and inner incision contamination. The introduction of PCR test to infection control practice is valuable in both clinical practice and scientific research. |
