Liver transplantation has been established as an effective treatment only for end-staged liver disorders. The acute and chronic rejection is also difficult to be deal with in clinical liver transplantation . Immunosuppressive therapy, including the use of cyclosporin and tacrolimus, is indispensable for successful liver transplantation , but these immunosuppressive drugs may promote the development of lethal complications. In order to reduce the dosage and time of immunosuppression, and make the recipient long survival, many experimental study has been made by learner all of the world. We practiced othotopic liver transplantation in rats by modified suture technique. Because hIL-10 is a potent regulator for controlling of allograft rejection, hIL-10 was conducted into liver of donor rats. We expected to prolong the survival of othotopic liver transplantation in rats and offer theorectical basis for inducing immunological tolerance and clinical organ transplantation.一,Studies on the ways of orthotopic liver transplantation in ratsIn order to controll two modified ways of orthotopic liver transplantation in rats. 30 cases of orthotopic rat liver transplantation have been performed using the improved three-cuff method of modulation of donor' s liver and modified cuffs. 70 cases of orthotopic rat liver transplantation have been performed using modified suture methods. The operative successful rate and one week survival rate for "cuff" methods were 80% and 8%, for modified "suture" methods 91% and 72% respectively.
