Experimental Study Of The Influencing Of Acupuncture On Ischemic Penumbra After Wistar Rats Focal Ischemia

The paper is directed at the study of the influencing of acupuncture on ischemic penumbra after rat focal ischemia and provides the theoretical basis for rescuing ischemic penumbra. The study involves the four methods (namely RT_PCR,TUNEL , Immunohistochemistry and TTC )to evaluate the CAD expression , neuronal apoptosis, expression of Bcl-2 and HSP70 protein in the IP and brain infarct volume, respectively.The experiment consists of three parts . The first part(experiment ) directes at the influence of acupuncture on the brain infarct volume of rats after MCAO(by TTC). The second directes at the influence of acupunct ure on CADmRNA expression in the IP(RT-PCR) . The third directes at the influence of acupuncture on neuronal apoptosis and the expression of Bcl-2 and HSP70 protein in the IP. Every experiment involved selection of 96 rats and the random division of them into four groups,namly normal control group(A,n=6),ischemic group(B,n=30), acupuncture group(C,n=30) and medication group(D,n=30). The latter three groups were subdevided into 3h ,6h, 12h, 24h and 48h groups.Results: Experiment One:Normal brain content was in red in A group;The infarct part was in white in B, C and D groups. Brain infarct volume of C group was smoller .than that of the B group(P<0.01);Experiment Two:Light CAD expressed in A group; In B group CAD was more significant than that of A group at 3h, peaked at 12h, and was meaningful at 48h; In C group CADmRNA was expressed lightly compared with B group(P<0.01);Experimernt Three:In A group no apoptosis cells, no Bcl-2 and HSP70 were expressed.
