Objective To investigate the method and its efficiency of transducingnm23-H1 gene into human bladder carcinoma T-24 cell line using electroporation in vitro, and to establish a stable , high-efficient way of transferring nm23-H1 gene into T-24 cells line.To evaluate whether nm23-H1 gene could affect the invasive ability and adherence ability and migration ability in T-24 cells line with transitional cell carcinoma of the bladder, and then to study the role of nm23-H1 gene in the improvement of sensitivity to anticancer agents in T-24 cells line.Methods The pCMV-Bam-neo-nm23-H1 plasmid was transferred intoT-24 cells line with Electroporation. The expressions of nm23-H1 gene between transferred and non-transferred T-24 cells line were detected by the method of immunohistochemistry and flow cytometric analysis (FCM). The invasion, adherence and migration ability between transferred and non-transferred cells line was detected by transwell-room and wash techniques. The chemo-sensitivity of T-24 cells line was evaluated by MTT.Results It was obtain the stable expression of nm23-H1 with pCMV-Bam-Neo system, and selected by using G 418.the significant difference of NDPKA expression between transferred and non-transferred T-24 cells line was discovered; The invasive ability, adherence ability and migration ability of transferred T-24 cells were decreased more tnan those of non-transferred T-24 (P<0.05), and the results of the non-transferred T-24 and the transferred T-24 show that the invasive ablilty was 65.60%±3.16%,33.93%±2.61% respectively(P<0.05), and the migration ablilty are 70.66%±3.81%, 47.95%±3.51% respectively(P<0.05). The chemo -sensitivity of transferred T-24 cells to Cisplatin (CDDP) increased significantly(P<0.05).Conclusion nm23-H1 gene can inhibit the metastasis of T-24 cells line and increase the chemo-sensitivity to CDDP significantly.
