Based on TCM theory and clinical experience , the prescription of TCM YIXINING of anti-stress-induced myocardium injury is made by our study . This study talks about effects and the mechanisms of Chinese herbs YiXINNlNG preventing and treating stress- induced cardiomyocytes injury in rat. Adopting restrainted- induced stress rat model, we tested the heart rates,ECG, apoptosis of cardiomyocytes and cellular phenotype in rat' s heart. Results: heart rates increasing, displacement of ST and highly degree upright of T wave in ECG, apoptosis of cradiomyocytes inducing, expression of fetal-type genes atrial natriuretic peptide inducing, protein synthesis at lweek increasing while decreasing at 4week in stress rats. After treated by pouring herbs YIXINNING down the throat, the YIXINNING group rats' heart rate decreased, the displacement of ST and upright degree of T wave in ECG improved significantly, numbers of cardiomyocytes apoptosis decreased sharply, gene expressions of ANP downregulated, protein synthesis in cardiomyocytes decreased significantly at 1st week , yet increased at 4th week. YIXINNING has remarkable protective effects on cardiomyocytes' injury caused by stress and is able to reduce the elevation of stress-induced plasma Cortisol significantly, upregulate the downregulation of stress- induced glucocorticoid receptor in rats. Further more it is able to lower the elevation of stress-induced serum catecholamine significantly, increase the downregulation of stress-induced β -adrenergic receptor in rats. The mechanism of YIXINNING improving the stress-induced myocardial ischemia proberbly related to it' s lowering the serum catecholamine and reducing toxic effects on catecholamine to myocardium. Its' anti-apoptosis effects maybe relate to restricting cell apoptosis pathway mediated byglucocorticoid receptor, and β1-adrenergic receptor. The mechanism of TIXINNING regulating cellular phenotypes could possibly relate to glucocorticoid-induced protein degradation and variation oC cell phenotype induced by β-adrenergic receptor.Above all, YIXINNING posses protective effect on strainted stress induced cardiomyocytes injuty in rats through regulating the contents of glucocorticoid and catecholamine and its receptors at different levels and targets.
