| We have investigated the mechanism of signal transmembrane transduction of cardiac hypertrophy induced by pressure overload,from gene transcription,protein expression to signal changes using the strech instrument of culture cell mimicking in vivo pressure overload. The main results as follows:1. According to document, a set of stretch instrument of culture cell has been developed from plexiglas and flexible membrane. It was proved that the instrument is simple and convenient to use.2. A new method has been established for cardiac microvascular endothelial cell culture, and its biological characters has been preliminarily studied.3. Many kinds of growth factors such as angiotensin â…¡ and endothelin secreted from fibroblast and microvascular endothelial cell induced by stretch will play an important role in stimulating cardiomyocytes hypertrophy.4. The cytoskeleton plays a role in the mechanism of cardiocyte hypertrophy induced by stretch through mediating the autocrine of cardiocytes after stretch.5. Specific extracellular matrix-integrin play a transmembrane signal transduction role in the mechanism of heart hypertrophy induced by pressure overload.6. Cardiocytes hypertrophic response stimulated by stretch could be inhibited by nitric oxide through decreasing the concentration of intracellulal free cacium. |
