| Objectives Isolation and culture murine mesenchymal stem cells(mMSCs);its abilities to suppress immunoreactive proliferation of splenic cells in vitro and to promote hematopoietic engraftment, decrease graft versus host disease (GVHD) of the recipients in allogenetic BMT were investigated. Methods LmMSCs were isolated with its adherence ability and purified by clonal culture. The biology and multiple differentiated abilities were studied. 2.In vitro , mMSCs suppressed the splenic cells proliferation induced by mitogen and alloantigen by means of MTT. 3.In vivo, the contribution of mMSCs to alleviate the clinical signs and pathology of GVHD in the recipients were observed, the impact on the T cells subset and cytokine secretion were also measured. Results 1.mMSC cell is purified , with renewal capacity and multiple differentiated abilities as cultured until 30 passage .2. mMSCs have immunomodulatory effects, can suppress splenic cells proliferative response stimulated by mitogen and alloantigen in dose-dependent manner.3. In vivo, mMSCs increase CD4~+CD25~+ regulatory cells and the secretion of anti-inflammatory cytokine , decrease the proinflammatory cytokine .They enhance hematopoietic engraftment and decrease GVHD of the recipients. Conclusions In BMT, mMSCs will be an important immunomodulatory factor for the prevention of GVHD and promotion of engraftment. |
