256 marine Streptomyces in JiaoZhou Bay were screened for their inhibitorybacteria activity. 10 typical strains were selected for chemical screening and theinteresting metabolites were isolated from 2 strains fermentation broth. The structure ofthe metabolites were analyzed. The significative 2 strains were classified according toculture characteristics, modality characteristics, characteristic reaction, and comparing16S rRNA gene sequence with other 5 strians.Staphylococcus aureus, Escherichia coli, Bacillus pyocyaneus, Sarcine sp.,Cryptococcus, Candida albicans, Mucor miehei (Tü 284), and Streptomycesviridochromogenes (Tü57) were used as testing strains, and the fermentation broth ofmarine Streptomyces were screened for inhibitory bacteria activity by liquid diffusionmethod. 22 % strains contain activity according to at least one test strain(inhibitiondiameter (?) ≥ 8 mm). The strains M024, M028, M042, M083, M086, M095, M097,M124, M134, and M226 were used to complete the chemical screening test on the basisof their modality characteristics and activity. 8 media and 4 culture conditions wereused to investigate metabolites generated by strains. Strains M095 and M097 werefound to be interesing strains. The optimum condition for M095 was Meat extractmedium, pH 6.5, 28℃, and 95 r/min. The optimum conditions for M097 were Meatextract medium, pH 7.8, 35℃, and 110 r/min(conditionⅠ)and M2+ medium, pH 7.8,28℃, and 95 r/min(conditonⅡ).Strains M095(24 L) and M097(30 L for conditionⅠ, 14 L for condionⅡ) werefermented and the metabolites were separated by ethyl acetate and columnchromatogram. The structures were analyzed by ESI-MS, EI-MS, 1H-NMR, 13C-NMR,and APTNMR. A high inhibitory compound Holomycin from M095 was found. It wasthe first time that we found Holomycin contains inhibitory fungal activity. Ten maincompounds were found from M097 and 8 compounds had different activities. The twoothers: Aloesaponaria Ⅱ was isolated from microorganism wild strain for the first time,and compound Cui D was a new anthraquinone compound.According to 16rRNA gene sequence analysis, the seven active marineStreptomyces were analyzed for their clustering relationship. They assumably belong to4 different group. Strain M095 maybe a variation speceise of Streptomyces griseus andstrain M097 maybe a distinct species within Globisporus group combining culturecharacteristics, modality characteristics, and characteristic reaction but need moreevidence.
