The Construction And Application Of Piezoelectric Cell-based Chip With Human Hepatoma Cell Line As Whole Biosensor

There are all kinds of recognized elements in a cell, such as receptors, ion channels and enzymes etc., which can be targets responding to varied stimulation and materials being analyzed. By acquiring and analying the response information from cells, the Piezoelectric Cell-based Sensor Chip (PCSC) was used to study cells or special materials. Plentiful information including growth and metabolism of cells, as well as responses to effects of complex and toxic materials can be obtained synchronously by the technology. Because of advantages in detection strategies such as being label-free, real-time, in-situ and non-invasive, this new analytical tecnology has been used widely in clinical diagnostics, environmental monitoring and food safety. It is most promisingly applicable for screening drugs, special of anti-cancer drugs, and is of significance in theoretical and practical research.The aim of the thesis is to construct the PCSC and find the response mechanic model of cell behavior in PSCS based on theory and experiments. By the technology the application in screening drugs and nano-cell biology were explored. Not only the task had promoted the analysis methods, but had it provided potential experimental proves for clinic medicine, as well as improvement of the nano-cell biology.The methods include combining the cell culture technology with a Piezoelectric Bio-Sensor Chip (PBSC, Homemade)instrument, a real-time analysis system established the PCSC system studying cancer cells behaviors. In order to verify the dynamic process of cell attachment behavior SEM and DTA assay were operated. During the application of PCSC MTT assay were used. At last HD method was used to prepare gold nanoparticles, which were proved by STM and UV photometer.Some achieved progresses have been obtained as follows:â‘ The analysis model of complexed cell system in PCSC was calculated as follow equations:It concluded the frequency shift was positive relative with impedence of acoustic wave.
