Objective: To find promoter sequence of Cryptococcusneoformans capsule——associated gene CAP10; To research on three carbohydratesand drugs' effects on activity of Cryptococcus neoformans capsule—associated gene CAP10 promoter。 Methods: constructing some recombinant plasmids that contain the different length of CAP 10 coding region 5 upstream flanking sequence by molecular clone method. Finding promoter sequence of CAP10 through two kinds of reporter gene system ; interfering transformant which was testified to have CAT expression activity with different concentrations of drugs and carbohydrates, then compare their influence on CAP10 promoter activity. Results and Conclusion: -303bp——0bp in CAP10 5' upstream flanking sequences act as a smallest intact promoter sequence. Glucose , mannose, fluconazole, amphotericin B and fluorocytosine have no induced effect on activity of CAP10 poromoter. Galatose has significant effect on CAP10 promoter and this effect is dose-dependent in the extension of experimental concentrations.
