Objective: To investigate the protective effects and mechanisim of Tongluo recipe on renal function in experimental diabetic nephropathy in rats. Methods: Diabetes was induced in mal SD rats by intrapetitoneal injection of STZ(60mg/kg). All rats were devided into 6 groups(TLR1,TLR2, MEL1, MEL2, CN, DM). Serium cholesterol level, urinary protein and blood glucose well detected as renal function index; glomerular area, PAS positive area, sirum red positive area, apoptotic cellular counts and histological changes in electron microscope were evaluated as histological damage index. The expression values of TGF-β1, GLUT1, JNK1, JNK2 and protein expression of JNK in renal cortex were quantitated with real-time PRC and immunohistochemistry. Results: Diabetic rats treated with Tongluo recipe or melatonin showed decreased urinary protein, moderately renal lesion, decreased gene expression of TGF-β1, GLUT1, JNK1, JNK2 and protein expression of JNK than those untreated rats. The correlated signal transduction in diabetic nephropathy is also impressed. Conclusions: Tongluo recipe and melatonin showed protective effect by decreased gene expression of TGF-β1, GLUT1, JNK1, JNK2 and protein expression of JNK in diabetic kidney.
