Ascertain of active compounds is a very important content in the development of traditional Chinese medicine. Due to its disadvantage of traditional research method of active compounds, the question of sample preparation and bioassay can't be resolved effectively and more attractive micro active compounds were ignored. The traditional Chinese medicine-Polygonum cuspidatum was selected as the research object in this paper, which has been identified to show potent estrogenic activity in MCF-7 cells. Preparative high performance liquid chromatography(P-HPLC)combined with recombinant yeast screening (YES) assay was employed in the process of studying the estrogenic active compounds in P. cuspidatum. The whole process was monitored by HPLC-UV analysis to research the correlation between the activity and the components.Based on the bioassay, the ethyl acetate fraction of ethanol extract of P. cuspidatum was isolated by silica gel plate and silica gel column chromatography respectively. Active components of column chromatography product were separated by P-HPLC secondly. Combining the results of HPLC analysis and estrogenic activity came to the conclusion that unknown bioactive compound might exist in the extract of P. cuspidatum. HPLC-MS analysis was used to identify this active compound. The results of primary research showed that the new activity component was likely to be citreorosein.
