BACKGROUND: esophageal carcinoma is a neoplasm of malignant epidermoid origin, one of the common malignant tumor in China,of which the etiology and mechanism remain unclear. The etiologic study showed correlation with trace element deficiency, nitroso-compound, and genetic and living habit. With futher investigation of oncogenes and tumor suppressor genes, biological studies on esophageal carcinoma were done,and provided the prognostic information. Proliferating cell nuclear antigen (PCNA) is an 36KD nuclear protein binding to DNA polymerase 5. and is associated with DNA replication.Recently,cyclinD1 has been identified on chromosom-e 11q13,and cyclinD1 protein had been classified as a G1 cyclin and played an important role in regulating the ceil cycle.Overexpression of cycIinD! may contribute to oncogenes by disturbing the cell cycle. DNA analysis for estimating the degree of aneuploidy of tumor cell is generally accepted as a valid parameter which demonstrates the malignant potentiality of tumors.The amplication and overexpression of PCNA and cyclinDl, and DNA content piayed an important role in development.progression and prognosis of esophageal carcinoma.MATERIALS AND METHODS:Samples were obtained from formalin— fixed , paraffin —embedded tissue blocks of 164 cases of esophageal squamous cell carcinoma. Immunohistochemologic-al staining for PCNA and cyclinD1,and in situ hybridization for cyclinD1 gene amplication, were performed on pre— and post —radiation specimens.
