Part one Clinical significance and detection of the C-erbB-2 proto-oncogene amplification in clinical samples of the baldder cancer by differential polymerase chain reactionThe C-erbB-2 proto-oncogene amplification was measured in clinical sample ( urine exfoliated cells, tumor tissue) of the 58 patient with transitional cell carcinoma. The target gene C-erbB-2 and control gene thymidine kinase are located on chromosome 17. The different size of two PCR fragments were produced by simulataneously performing PCR on test and control genes in the same PCR mix. Then, Two PCR fragments were separated by 2% Nusieve GTG agarose gel electrophoresis. The densitometry was used to measure the ratio between C-erbB-2 and thymidine kinase. The C-erbB- 2 proto-oncogene amplification show that the ratio is higher than 1. Results: No The C-erbB-2 proto-oncogene amplifiation was identified in the normal healthy individuals and patients of the non-tumor-bearing bladders. Grade I , II and III showed the C-erbB-2 proto-oncogene amplification in 0/16 (0%), 7/28 (25%) and 8/14 (56%)...
