Background: Alzheimer's Disease( AD) was reported by Alois Alzheimer in 1906. AD is characterized by progressive degeneration of the brain. It showes a variety of pathologic characters including neurofibrillary tangles, neuritic paques and degeneration of central cholinergic neuron. AD is caused by multiple factors, such as environment, genetics, immunity, apoptosis, cytokines and so on. There are more than 5 genes related to AD genetic susceptibility including Apolipoprotein E (ApoE), amyloid protein precursor (APP), presenilin-1 (PS-1), presnilin-2 (PS-2) and α1-antichymotrypsin (ACT) genes. Objective: To study the association of Alzheimer's disease(AD) with the apolipoprotein E (ApoE) e 4 allele and 1/1 genotype of presenilin-1 in Chinese and to find the biolobical peripheral makers for the laboratory diagnosis of AD; To explore the mechanism of presenilin mutation causing Alzheimer's disease and to provide the evidence for the prevention and treatment of AD. Material and Methods: Seventy-four patients with AD, male 65, female 19, average age of 74.05±10.44 (mean±SD); 56 patients with vascular disease (VD), male 43, female 13, average age of 73.36±6.95 (mean±SD); 78 age-matched non-demented individuals, male 62, female16, average age of 71.39±7.70 (mean±SD) are collected.
