Lung cancer cells can upregulate the expression of Fas ligand (FasL) and counterattack tumor-infiltration lymphocyte (TIL) expressing Fas via the FasL/Fas pathway, therefore escaping from immunosurvillance and impairing local anti-tumor immune capacity. As so, FasL is a promising target in gene therapy against lung cancer. Alternatively, tumor mesenchyme provides important cues for invasive growth of tumor cells, in which rapid proliferation of mesenchymal cells is required. Being precursor for all mesenchymal lineages, mesenchymal stem cells (MSC) can migrate toward cancer bed or tissue surrounding the tumor, known as'chasing down'infiltration phenomenon, participating in and enhancing the formation of tumor stroma through proliferation as well as differentiation. MSC has been isolated from bone marrow (BM), liver, spleen, muscle, brain, skin and fat. However, it is rare known about its distribution in lung. Moreover, several key questions regarding lung-derived MSC remain poorly understood, including its immunoregulatory effect, and the correlation with ontogeny, development and metastasis of lung cancer, which illustration will undoubtedly further the appreciation of interaction between lung cancer mesenchyme and stem cells. The following is a brief summary of our research.1. Identification of Fetal Lung-Derived Mesenchymal Stem CellsFetal lung tissue was obtained from fetus of 15-22 weeks pregnancy of voluntary donation to confirm the existence of lung-derived mesenchymal stem cells. As a first step, mononuclear cell (MNC) was isolated by density gradient centrifugation, then expansion of fetal lung-derived adherent cell (FLAC) with low serum culture system. Subsequently, separate clones were individually plucked for further amplification and identification. Immunophenotype was decided by flow cytometry, and the general biology features were determined in several aspects including cell morphology, cell cycle and growth curve.
