PART ITHE STUDY OF MECHANISM ON GSP INDUCING GBC-SD CELL LINE APOPTOSISObjective Gallbladder cancer is a dismal disease with low response to chemotherapy, radiotherapy and immunotherapy. Therefore, new treatment modalities are important to effectively control and palliate gallbladder cancer disease.In this study, we investigate the effect of GSP on the growth and apoptosis of GBC-SD cell lines and illustrate its possible mechanism.Methods GBC-SD cell lines were exposured to different concentrations of GSP, the cytotoxicity was detected by MTT assay, the rates of apoptosis were examined by flow cytometry. The changes of cell morphology were obsered by light microscopy and the transmisson electron microscopy. DNA ladder assay was employed for the apoptosis. Flow cytometry was used to detected the changes of P53 , Bcl-2.Rusults The growth of GBC-SD cell lines could be significantly inhibited by GSP with concentration and time dependente. Typical apoptosis change was observed in 72h by light microscopy and the transmisson electron microscopy, including cell shrinking, chromatin condensation and apoptotic body. There were characteristic ladder bands after treated by GSP(3μg/ml, 6μg/ml) for 72h.The apoptosis rate investigated by flow cytometer was0.23 ±0.02%, 8.01 ± 0.08%, 13.27 ±0.12%, 22.07± 0.23%, 36.59±0.37%, respectively at concentration 0μg/ml,...
