Fresh Houttuynia cordata Thunb. is a Chinese materia medica that generally used in the Chinese medicine therapy. It possesses the actions of clearing heat, eliminating toxins, reducing swelling, discharging pus and relieving stagnation. However, most of the previous related studies were mainly focused on the essential oil. The common used quality standard of H. cordata is still relied merely on the undecanone that served as a marker compound for the qualitative evaluation standard as stipulated in the Chinese Pharmacopoeia (2005 edition). But undecanone alone could not be responsible for the overall pharmacological activities of H. cordata and synergistic effects between various constituents may exist. The chemical studies were mainly focused on the liposolubility compounds. So we systematically study the chemical of H. cordata combination with the pharmacodynamics. And establishment of GC-MS and HPLC-DAD-MS fingerprint of fresh H. cordata . The fingerprints were used to differentiate among different portions of herb, difference in processing methods, between dry and fresh medicine.The fresh H. cordata were extracted three times with methanol, the extract was concentrated under reduced pressure to a small volume. The concentrated liquor was subjected to solvent-solvent partitioning with ether, chloroform, ethyl acetate, and n-butanol respectively. Each part was concentrated under reduce press. In this study, the chloroform, ethyl acetate and n-butanol extract were segregated over Diaion HP-20 , Sephadex LH-20, ODS and silica gel column repeatedly, 24 compounds were isolated and purified. On basis of spectral and chemical evidence such as ESI-MS, 1HNMR, 13CNMR, 1H-1HCOSY, HMQC, HMBC, DEPT etc., structures of 19 compounds were elucidated. Among these compounds, 9 compounds were separated from H. cordata for the first time, such as HC-16 (Kaempferol-3-O-β -robinobinoside ), E-2-3(quercetin-3-O- β -D-galactoside-7- O-β -D-glucopyranoside), HC-31 (quercetin 3-O-α-L- rhamnopyranosyl -7-O-β -D-glucopyranoside), HC-32 ((E)-4- Hydroxy -4- [3'-( β -D- glucopyranosyloxy) butylidene] —3,5,5—tri methyl -2-cyclohexen-l-one), HC-34(2-(3,4-dihydroxy) -phenyl-ethyl- β -D- glucopyranoside), HC-W-2 (p-hydroxyphenethyl- [ β -D-glucoside]), HC-2(chlorogenic methyl ester), HC-9(4-( β -D-glucopyranosyloxy)-3-hydroxy- Benzoic acid), HC-38(KNO3). Besides, HC-7(hyperin), HC-8(quercitrin), HC-14(rutin), HC-E-5(isoquercitrin), HC-39(quercetin), HC-47(piperolactam A), HC-12(3,4-dihydroxybenzoic acid), HC-M(chlorogenic acid), HC-50(Aristololactam B) and HC-52(Cepharadione B)...
