BackgroundThe abnormalities of serum lipids are associated with the development of coronary heart disease (CHD).The apolipoproteins play an important role in lipid metabolism, the change of their molecular structure or their expression level significantly changes serum lipids. The abnormality of serum lipid is the major risk factor of atherosclerosis and CHD, and it also increases the susceptivity of hypertension, obesity and insuline resistance.ApolipoproteinA5(ApoA5) is a newly identified apolipoprotein by comparative human and mice genome which located at the vicinity of ApoAl/C3/A4 gene cluster, ApoA5 was found to be significantly negatively associated with plasma triglyceride levels in animal studies. Single nucleotide polymorphism (SNP) of ApoA5 had shown the effect on lipid metabolism. Some studies linked ApoA5 to HDL and LDL. Recently it has shown that ApoA5 mediated inflammation reaction. These findings indicate that ApoA5 may be a major risk factor for coronary artery disease. However, little is known about that the concentration of serum ApoA5, and whether ApoA5 SNP modulates the lipid metabolism through function transform or content. The relationship between ApoA5 and serum lipids and CHD are not clearly elucidated. Furthermore, ApoA5 can not be isolated directly from serum for its low content. This study is to express recombinant protein with gene engineer, to produce antibody of ApoA5, and to detect the level of ApoA5 between healthy controls (n=134) and CHD subjects (n=73). ObjectiveThe study was to construct the prokaryotic expression vector to prepare the infusion ApoA5 and the antibody to ApoA5. Second, detect the level of ApoA5 in serum and various tissues. Third, investigate the association of ApoA5 with serum lipids and CHD.
