| Repairing articular cartilage defect has been a difficult task because of the poor ability of intrinsic cartilage repair. But this problem is not solved finely tody. Traditional methods such as microfractrue, abrading, periosteum or perichondrium or osteocartilage replacement, prosthetic replacement have some disadvantages. In recent years, there has been major advance in tissue engineering, which is expected to provide a new treatment method for repairing cartilage defect. Tissue engineering is an interdisciplinary science of co-development and combination of modern cell biology, biomaterial science and engineering. The establishment of tissue engineering is a breakthrough over conventional organ transplantation which restore the function and structure of tissues lost in the injury and disease. Tissue engineering, embryonic stem cell and genetic engineering are regarded as the three marvels in the history of life science development. As so far, the problems need to be resolved in tissue-engineered cartilage application are as following: â‘ a source of pluripotentional cells with strong ability of proliferation and differentiation; â‘¡ an appropriate cells' carrier; Therefor, the present study is designed and contains three main parts.Chapter I Experimental study of the isolation, culture and biological characteristics of rat adipose derived mesenchymal stem cellsObjective To master method for the isolation culture and amplification of rat adipose derived mesenchymal stem cells(ADMSCs) in vitro and explore their biological characteristics. Method The inguinal fat pads were excised with sterile technique, followed by digestion with 0.075% collagenase, then the cells were cultrued in Dulbecco modified Eagle medium with 10% new-born calf serum. The morphology of ADMSCs was observed with inserted microscope constantly. Passage cells were examined using flow cytometric and immunohistochemical methods about CD antigens: 29, 34, 44.
