| Alzheimer disease (AD) is a neuro degenerative disease with clinical feature of memory impairment, cognitive disorder and neuro functional impairment. It is a senile disease associated with aging of body and age. The pathologic feature of AD are neuro fibrillary tangles (NFT) which is in never cells and senile plaque(SP)s which is out of never cells and mainly at frontal lobe and hippocampus. The etiological factors and pathogenesis of AD are unknown, and there is no effective therapy method, therefore more deeply studies on it are still needed. At present, there is no ideal animal models simulating all aspects of AD , it limeted the basic studies of AD and development of drugs on AD. so it is an earger to find an ideal and applied animal models of AD; Among the etiological factors, eld and deposition of β- amyloid(Aβ) in brain tissues are main causes of AD, so the biological change of AD in eld to be approached; there are many traditional Chinese medicine compound recipes on AD which major components are Polygonum multiflorum(PM), and PM is demonstrated to be a drug on AD, so the mechanism of PM on AD stay to be elucidated accurately.I The study of memory disorder in old rats with ADObjective: To set up the AD model by healthy old rats with obvious memory disorder.Methods: SD rats were supplied by experimental animal centre of Central South University (CSU) . "Y" shape maze test chamber was made by ourselves according to references. Firstly, 40 old (20 months) rats were tested and the 10% of them had worst memory which needed at least 34 times of electric shocks. Hence, 15 old rats which needed at least 34 times of electric shocks were selected as the memory disorder group(MDG), 15 old rats which needed at most 33 times of electric shocks were selected as the ordinary old group(OOG), 15 young(2 months) rats were selected as the young group(YG). All rats were tested by maze test, then the mices were decapitaed and the hippocampus tissues were extracted for the test of mRNA of APP cleaving enzymes (APPCE)... |
