Source: Province natural science fund of Guangdong (23003) and province science and technology of Guangdong offend to close the item (2KM05101S). Purpose and meaning: 1. Look for a kind of method to check the expression of immune activation genes in lymphoid cell, which meet for clinical demand, high in sensitivity, strong in specificity, efficiently and economical; 2. Study the relation between the expression of immune activation genes in lymphoid cell and the acute rejection after liver transplantation by the method affirmed before. To explore the sensitive genes that can predict the occurrence of acute rejection after liver transplantation. Method: 1. To select the best method which most meets for the clinical demanding at the sensitivity, specificity, efficiently and economical. 2. To study the relation between the expression of immune activation genes in lymphoid cell and the acute rejection after liver transplantation by the method affirmed before. Contents and processes: 1. 14 liver transplantation receptors (7 receptors were of acute rejection, the others not) were taken 15ml blood, which was divided into three equal parts. Reverse transcription polymerase chain reaction (RT-PCR), Taq man probe fluorogenic PCR and SYBR Green I real-time quantitative PCR were used to detect the expression of the immune activation genes.
