| Objective Alcoholic liver disease (ALD) is a public and topic health problem all over the world. Histopathological features of ALD include adiposis hepatica , steatohepatitis , followed by fibrosis and cirrhosis . Now the etiopathogenesis of ALD remains obscure. In recent years, savas presents with the hypothesis of two hits which is characteristic by center of oxidase stress and lipid peroxidation to account for the mechanism of alcohol induced liver injury. The first hit causes lipid deposition in the hepatic cells which leads to adiposis hepatica. The second hit involves oxidase stress and lipid peroxidation. Endotoxin and proinflammatory cytokines are two inseparated main parts to take part in the process. They can activate chuffer cells which are resident hepatic macrophages. The activated chuffer cells (KC)overexpress NADPH oxidase(NOX) and cyclooxiygenase-2(COX-2), and they also produce oxygen free radical and cytokines to recruit neutrophilic leukocytes and lympholeukocytes's infiltration which result in inflammation to take part in the etiopathogenesis of ALD.The aim of present experiment is to develop a rat model of ALD by intragastric alcohol. The contents of liver enzymes and cytokines in serum and the index of lipid peroxidation in the liver homogenate are examined dynamic. The expression of kupffer cells , NADPH oxidase, cyclooxiygenase-2 and Toll like receptor 4 in liver tissues is assessed. We combined those results with the pathology of hepatic tissue to explore the etiopathogenesis of ALD from macro-or microscopy and molecular level , and to investigate the therapeutic and preventive effect of Zhiganning Chinese herbal compound on ALD, and to elicit the action mechanism of the medicine... |
