| As a prevalent clinic symptom of many cardiovascular disease, Cardiac hypertrophy is intensively related with the mobidity of heart failure, atherosclerosis and stroke. Recently isolation of disease related genes has been an important field in revealing the pathogenesis of various disease, samely, studies on related genes of cardiac hypertrophy can also bring us a useful clue in understanding the triggers and signaling mechanisms determining those changed relevant phenotypes. In our group's previous research, we have used cDNA-substractive hybridization method to isolate the related gene fragments in rat models of pressure-overload cardiac hypertrophy, there are 36 fragments isolated and all show significant differences between the control and model materials by clony and dot hybridization with cDNA probes. Sequence homogenous comparing shows that some of them are similar to known genes, such as cytochrome b, creatine kinase, rab 11 b etc. The SMART RACE method was applied to get the full-length cDNA sequence of creatine kinase (1479 bp) and cytochrome b (1172 bp), including complete reading frame with the start code and the stop code. Then the full length cDNA was cloned into PGEM-T vectors and identified by sequencing, The results of Northern hybridization analysis have also confirmed the significant differences between the control and model materials . Becuse creatine kinase and cytochrome b are involved in the energy metabolism of cardiomyocyte, it shows that they could be... |
